摘要
目的探讨胃黏膜水通道蛋白3(AQP3)、水通道蛋白4(AQP4)表达与慢性胃炎的相关性。方法选取2015年1月至2016年12月在该院治疗的慢性胃炎患者72例,其中慢性非萎缩性胃炎(CNAG组)患者47例,慢性萎缩性胃炎(CAG组)25例,同时选取30例正常胃黏膜组织作为对照组,采用免疫组化染色法检测AQP3和AQP4蛋白表达,RT-PCR检测AQP3和AQP4 mRNA表达,同时观察治疗前后慢性胃炎患者AQP3和AQP4蛋白和mRNA表达情况。结果 CNAG组AQP3和AQP4 mRNA表达分别为(0.782±0.103)×104、(0.430±0.073)×104 copies/μg RNA,蛋白的表达分别为0.803±0.101、0.501±0.084,明显高于CAG组和对照组(P<0.05);CAG组AQP3和AQP4mRNA表达分别为(0.432±0.099)×104、(0.211±0.080)×104 copies/μg RNA,蛋白的表达分别为0.572±0.095、0.317±0.080,明显低于对照组(P<0.05)。CNAG组治疗后AQP3和AQP4 mRNA和蛋白表达均较治疗前降低(P<0.05);CAG组治疗后AQP3和AQP4mRNA和蛋白表达均较治疗前升高(P<0.05)。结论 CNAG和CAG胃黏膜中AQP3、AQP4mRNA和蛋白表达有所不同,治疗后均有所改善,AQP3、AQP4的表达异常可能与胃炎有一定的关系。
Objective To investigate the correlation between the expression of gastric mucosal aquaporin 3(AQP3)and aquaporin 4(AQP4)and chronic gastritis.Methods 72 patients with chronic gastritis treated in the hospital from January 2015 to December 2016 were selected,including 47 cases of chronic non-atrophic gastritis(CNAG group),25 cases of chronic atrophic gastritis(CAG group),and 30 normal gastric mucosa tissues were taken as the control group.The expressions of AQP3 and AQP4 protein were detected by immunohistochemical staining,and mRNA of AQP3 and AQP4 protein was detected by RT-PCR.The expression of AQP3 and AQP4 protein and mRNA in patients with chronic gastritis before and after treatment were observed.Results The expressions of AQP3 and AQP4 mRNA in group CNAG were(0.782±0.103)×10^4 and(0.430±0.073)×10^4 copies/μg RNA respectively.The expressions of protein were 0.803±0.101 and0.501±0.084,which were higher than those in CAG and control groups(P〈0.05).The expressions of AQP3 and AQP4 in CAG group were respectively(0.432±0.099)×10^4 and(0.211±0.080)×10^4 copies/μg RNA.The protein expressions were 0.572±0.095 and 0.317±0.080,which were significantly lower than those in the control group(P〈0.05).The expressions of mRNA and protein of AQP3 and AQP4 in CNAG group were lower than those before treatment(P〈0.05).The mRNA and protein expressions of AQP3 and AQP4 in CAG group were higher than those before treatment(P〈0.05).Conclusion The expressions of mRNA and protein of AQP3 and AQP4 in the gastric mucosa of CNAG and CAG were different,and all improved after treatment.The abnormal expressions of AQP3 and AQP4 may have a certain relationship with gastritis.
