摘要
目的:研究妊娠期高血糖对雄性子代成年以后肝脏脂质代谢的影响,初步探讨长链非编码RNA(lncRNA)肝脏特异性甘油三酯调节剂(LSTR)的调控机制。方法:采用8周龄健康成年雌性SD大鼠,糖尿病模型组20只,对照组10只,分别与健康雄鼠交配。交配成功后,糖尿病模型组腹腔注射35mg/kg链脲佐菌素溶液,对照组腹腔注射柠檬酸-柠檬酸钠缓冲液。两组雄性子代分别记为糖尿病组子代和对照组子代。全自动血生化分析仪检测血脂,qPCR检测肝脏基因表达。结果:糖尿病组子代比对照组子代表现高出生体重(t=3.193,P=0.020)、生长趋势体重明显增加和血甘油三酯升高(t=2.1970,P=0.0310)。相对于对照组子代,糖尿病组子代组Apoc2表达降低(t=10.20,P<0.001),lncRNA LSTR表达升高(t=12.29,P<0.0001)。糖尿病组子代Apoc2表达与lncRNA LSTR表达呈负相关(R2=0.2582,P=0.0008)。结论:母亲GDM可能是子代成年期发生肝脏脂质代谢性疾病的一个高风险因素。
Objective: To investigate the effect of maternal gestational diabetes mellitus on SD rat liver lipid metabolism and the lncRNA LSTR expression. Methods: Sprague-Dawley rats( 8-week-old) were used in this study,20 rats in GDM group,and 10 rats in control group.Maternal gestational diabetes mellitus was induced by a single injection of streptozotocin on the first day of gestation.Control rats were injected with citrate buffer.Serum lipids were determined by Automatic blood biochemical analyzer. Gene expression were evaluated through qPCR in islet.Results: Compared with control group offspring,diabetes group offspring showed more higher birthweight( t = 3. 193,P = 0. 020),body weight and triglycerides( t = 2. 1970,P = 0. 0310). In addition,the Apoc2 expression decreased in diabetes group offspring( t = 10.20,P〈0.001) and the lncRNA LSTR expression increased( t = 12. 29,P 〈0. 0001). The expression of Apoc2 was negatively correlated with the expression of lncRNA LSTR( R2= 0.2582,P = 0.0008). Conclusion: Maternal gestational diabetes mellitus may be a high-risk factor of liver lipid metabolism disturbance in male offspring.
