摘要
目的:探讨ALK2在小鼠牙胚中的表达及牙胚标本和切片的制备方案。方法:取E12.5-E18.5的小鼠胚胎,4%多聚甲醛于4℃固定24 h后,按脱水总时间23 h和8.5 h两种方案脱水,石蜡包埋切片后,HE染色观察标本制备情况,免疫组织化学染色观察ALK2在牙胚中的表达情况。结果:大体显微镜下观察到的胚胎形态和文献所述一致。脱水总时间8.5 h的脱水方案所制备的石蜡切片平整、完整、无裂痕,染色质量良好,牙胚形态完整,结构清楚。免疫组化染色发现,ALK2广泛地表达于蕾状期的口腔上皮和间充质,帽状期及钟状期的成釉器、牙乳头。结论:减少脱水时间,不影响切片和染色质量,可提高实验效率。ALK2广泛表达于牙胚早期,可能在牙发育中具有重要作用。
Objective: To investigate the expression of ALK2 in mouse tooth germ and the preparation of tooth germ specimens and slices. Methods: The mouse embryos of E12. 5-E18. 5 were fixed with 4% paraformaldehyde at 4 ℃ for 24 h and dehydrated for the total time of 23 h and 8. 5 h respectively according to different dehydration schemes. After paraffin embedded section,HE staining was used to observe the specimens. The expression of ALK2 in the tooth embryo was observed by immunohistochemistry. Results: Embryo morphology observed under gross microscope was consistent with the literature. The paraffin sections prepared by the modified 8. 5 hdehydration sheme were smooth,complete,no crack,with good dyeing quality,complete morphology and clear structure. Immunohistochemical staining showed that ALK2 was widely expressed in the oral epithelium and mesenchyme of the bud stage,the enamel organ and dental papilla of the cap and bell stage. Conclusion: Reducing the time of dehydration does not affect the quality of slicing and dyeing,improves the experimental efficiency. ALK2 is widely expressed in the early development of teeth,suggesting that ALK2 may play an important role in tooth development.
