摘要
目的分析焦炉工外周血中DNA甲基化转移酶1(DNMT1)、组蛋白去乙酰化酶1(HDAC1)蛋白的表达情况,探讨焦炉工人肺癌筛查的生物标志物。方法以152名男性焦炉工人为接触组,以141名男性水处理工为对照组。采集2组人员晨尿,用高效液相色谱法检测其内暴露指标1-羟基芘(1-OH-Py)的水平。同时采集晨起空腹静脉血,采用酶联免疫吸附试验(ELISA)检测血清中DNMT1、HDAC1蛋白的表达,并分析DNMT1、HDAC1蛋白的表达与焦炉工人工龄、工种、吸烟、饮酒等的关系。结果接触组尿1-OH-Py水平[(0.043±0.011)μg/mmol Cr]和血清中DNMT1[(28.4±7.2)μg/L]、HDAC1蛋白[(382±64)μg/L]的表达高于对照组[(0.017±0.004)μg/mmol Cr、(17.6±3.0)μg/L、(246±25)μg/L],差异有统计学意义(P<0.001)。接触组中不同工种间DNMT1、HDAC1蛋白表达水平也不同,差异有统计学意义(P<0.05)。随着焦炉作业工龄的增加,接触组DNMT1、HDAC1蛋白表达水平都有逐渐升高的趋势,差异有统计学意义(P<0.001)。结论血清中DNMT1、HDAC1蛋白可作为焦炉工人肺癌早期筛查的生物标志物。
Objective To determine the expression of DNA methyltransferase 1（DNMT1） and histone deacety-lase 1（HDAC1） protein in the peripheral blood of coke oven workers, and to investigate the biomarkers of lung cancer screening in coke oven workers. Methods A total of 152 male coke oven workers were included in the exposure group, and 141 male water treatment workers were included in the control group. The morning urine in the two groups was collected, and the level of 1-hydroxy pyrene（1-OH-Py） was examined by high performance liquid chromatography（HPLC）. At the same time, the fasting vein blood was collected in the morning. The expression of DNMT1 and HDAC1 protein in serum was determined by enzyme-linked immunosorbent assay（ELISA）. The relationship between the expression of DNMT1 and HDAC1 protein and working age, type of work, smoking and drinking of the coke oven workers were analyzed. Results The level of urinary 1-OH-Py and serum DNMT1 and HDAC1 protein expression in the exposure group were higher than those in the control group [（0.043 ±0.011） μg/mmol Cr vs（0.017 ±0.004） μg/mmol Cr,（28.4±7.2） μg/L vs（17.6±3.0） μg/L,（382±64） μg/L vs（246±25） μg/L],and the differences were statistically significant（P〈0.001）. The expression levels of DNMT1 and HDAC1 protein were different among the workers with different types of work in the exposure group, and the differences were statistically significant（P〈0.05）. With the increase of the working age of coke oven work, the expression levels of DNMT1 and HDAC1 protein in the exposure group showed a trend of gradually increasing, and the differences were statistically significant（P〈0.001）. Conclusion Serum DNMT1 and HDAC1 protein can be used as biomarkers for early screening of lung cancer in coke oven work-ers.
