摘要
目的研究肾透明细胞癌中SLAMF1基因的表达变化,评价SLAMF1基因表达与临床病理学特征的相关性及其诊断预后价值。方法利用oncomine和TCGA数据库的RNA-Seq数据和临床数据,分析SLAMF1基因表达与肾透明细胞癌患者临床病理学特征及生存预后的关系。结果 oncomine数据库的Yusenko Renal、Beroukhim Renal、Jones Renal队列以及TCGA数据库中肾透明细胞癌组织(180例)和正常组织(111例)表达谱数据分析表明,SLAMF1基因在肾透明细胞癌组织中的表达均显著高于正常组织(P<0.05)。分析524例肾透明细胞癌患者临床病理学特征与SLAMF1基因表达的关系显示,SLAMF1表达与Grade分级(P<0.001)、T分期(P<0.01)、M分期(P<0.01)和临床分期(P<0.001)均有显著性差异;生存分析结果显示SLAMF1基因高表达患者的总生存时间显著短于低表达患者(Log-rank=8.738,P<0.01)。ROC曲线分析结果表明,SLAMF1基因表达是一个灵敏度和特异度较高的肾透明细胞癌诊断指标(AUC=0.809)。结论 SLAMF1高表达与肾透明细胞癌的发生、发展相关,是候选的癌基因。SLAMF1基因的表达对判断肾透明细胞癌的生存预后及其诊断具有参考价值。
Objective To explore the expression of signaling lymphocytic activation molecule family member 1（ SLAMF 1） in renal clear cell carcinoma（RCCC） and its relationship with clinicopathological characteristics and prognosis of RCCC. Methods The RNA-Seq and clinical data in the oncomine and TCGA databases were analyzed. Results The Yusenko Renal,Beroukhim Renal and Jones Renal cohorts in the oncomine and TCGA databases（including 180 tumor tissues and 111 paracancerous tissues） revealed that the expression of SLAMF 1 was significantly higher in RCCC tissues than in healthy tissues（ P 〈0.05）.The expression of SLAMF 1 was significantly correlated to Grade classification（ P 〈0.001）,T stage（ P 〈0.01）,M stage（ P 〈0.01） and clinical stage（ P 〈0.001）.Survival analysis demonstrated that the overall survival of RCCC cases with high expression of SLAMF 1 were shorter than those with low expression（ Log-rank =8.738, P 〈0.01）.ROC curve analysis illustrated that SLAMF 1 expression was a diagnostic marker of RCCC with good sensitivity and specificity（ AUC =0.809）. Conclusion The high expression of SLAMF 1 is associated with the occurrence,invasion and metastasis of RCCC.The SLAMF 1 expression is helpful in the diagnosis and prognosis of RCCC.
