摘要
为了探究宿主蛋白Hsp90AB1在狂犬病病毒(Rabies virus,RABV)复制过程中的功能,以RABV疫苗毒株HEP-Flury感染鼠神经瘤母细胞N2a为模型,分析Hsp90AB1对RABV复制的影响。采用RNA干扰的方式,发现Hsp90AB1基因敲降后可影响病毒基因的转录及其表达,而未影响病毒的反基因组水平以及病毒感染力,表明Hsp90AB1在转录或蛋白表达水平上影响RABV的复制。由此初步发现Hsp90AB1在RABV复制过程中的作用方式,为继续深入研究Hsp90AB1在RABV感染过程中的作用机制提供了基础数据。
In order to investigate the function of host protein Hsp90AB1 in rabies virus(RABV)replication,the effect of Hsp90AB1 on RABV replication was analyzed preliminarily by using the model of mouse neuroblastoma N2 acells infected with rabies virus(RABV)vaccine strain HEP-Flury.Using RNA interference,it was found that Hsp90AB1 knockdown affected the transcription and expression of viral gene,but did not affect the level of viral anti-genomic RNA and virus infectivity,indicating that RABV replication was affected by Hsp90AB1 at the protein level or at the transcription level.The above results showed preliminarily the effect of Hsp90AB1 on RABV replication,which provided the basic data for further study on the detailed mechanism of Hsp90AB1 during RABV infection.
