摘要
目的:探究水蛭经炮制后的化学成分变化情况。方法:将一批水蛭作为实验对象,对水蛭实施高效液相色谱法(HPLC)-柱后衍生化法炮制,对经过炮制的水蛭中的氨基酸成分予以测定,并对甾体类化合物组成采用气相色谱-质谱联用仪(GC-MS)测定,对炮制前后氨基酸及甾体类化合物的成分变化情况予以比较分析。结果:经过炮制后的水蛭,其氨基酸含量如天冬氨酸、苏氨酸、谷氨酸等成分与炮制前比较明显降低,氨基酸总量由原来的73.93%降低到45.04%,差异存在统计学意义(P<0.05),甾体类化合物组成成分也发生明显变化,胆固醇氧化产物呈现出明显的增加趋势,差异统计学有意义(P<0.05)。结论:水蛭经过炮制后,蛋白质空间构象将会遭到破坏,使氢键断裂,促使氨基酸与甾体类化合物氧化分解,降低氨基酸成分,使甾体类化合物组成成分发生改变,需加强临床关注。
objective:to investigate the chemical composition changes of leech after processing.Methods:our new purchases a batch of Chinese medicine leech as experimental object,the leeches implementation of high performance liquid chromatography(HPLC)-column derivatization method after processing,and after processing of leeches in the amino acid composition was to be measured,and the nonsteroidal compounds by gas chromatography/mass spectrometry(GC-MS)determination of the chemical composition of amino acids and nonsteroidal compounds before and after changes to the comparative analysis.Results:after processing the leeches,and its amino acids such as glutamic acid,aspartic acid,threonine,the composition such as significantly lower compared with before anything,decreased from 73.95% to 73.93%,and the total amino acid differences there was statistical significance(P〈0.05),nonsteroidal compound composition also had obvious changes,cholesterol oxidation products increased obviously statistically significant(P〈0.05).Conclusion:leeches after processing,protein conformation will be destroyed,the hydrogen bond rupture,prompt amino acid and nonsteroidal compound oxide decomposition,reduce the amino acid composition,make the nonsteroidal compound composition change,need to strengthen clinical attention.
