摘要
目的:研究野艾蒿总黄酮的提取方法及其对人肝癌细胞(HepG2)细胞脂代谢的影响。方法:采用乙醇洗脱大孔树脂吸附柱的方式提取野艾蒿中的总黄酮,通过紫外分光光度法检测样品中总黄酮的含量。以HepG2细胞作为研究对象,MTT法检测野艾蒿总黄酮对HepG2细胞增殖的影响。制备HepG2细胞脂质堆积模型,油红0染色OD值法和试剂盒法观察并检测野艾蒿总黄酮对该模型中脂质代谢的影响,以及总胆固醇、甘油三酯含量的影响是否通过腺苷酸蛋白活化激酶(AMPK)得以实现。结果:野艾蒿中总黄酮的含量为44.56 mg/g,且可不同程度地抑制HepG2细胞增殖(P<0.05),药物浓度为0.010 g/L且用药24 h为最适实验条件;野艾蒿总黄酮可显著改善HepG2细胞中的脂质堆积情况,并下调总胆固醇和甘油三酯的含量(P<0.05),但此种作用可被AMPK抑制剂所阻断。结论:野艾蒿总黄酮可通过调节AMPK这一靶点起到调节HepG2细胞内脂质代谢的作用。
Objective： To study the extraction method of the total flavones in Artemisia lavandulaefolia and its effect on the lipid metabolism in HepG2 cells. Methods： The total flavonoes in Artemisia lavandulaefolia were extracted by adsorption columns of ethanol elution macroporous resin,and the total flavonoes in the samples were detected by ultraviolet spectrophotometry. HepG2 cells were taken as the research object,with the influence of the total flavonoes in Artemisia lavandulaefolia on proliferation of HepG2 cells determined by the MTT method. A HepG2 cell lipid accumulation model was prepared,and the oil red 0 stained OD value method and the kit method were used to observe and test whether the effect of total flavones in Artemisia lavandulaefolia on lipid metabolism and total cholesterol and triglyceride content in this model was achieved by AMPK. Results： The content of total flavones in Artemisia lavandulaefolia is 44.56 mg/g,which can inhibit the proliferation of HepG2 cells to different degrees（P〈0.05）. But this effect can be blocked by AMPK inhibitors. Conclusion： Total flavones in Artemisia lavandulaefolia plays a role in regulating the lipid metabolism in HepG2 cells by regulating the target of AMPK.
