口腔牙托水对大鼠细胞色素P450酶活性的影响 被引量:1

Effects of methyl methacrylate on CYP450 in rats

摘要 目的研究牙托水对大鼠CYP1A2、CYP2B6和CYP2C19酶活性的影响。方法 24只雄性SD大鼠,随机分为正常对照组、灌胃牙托水3 d组、灌胃牙托水10 d组。牙托水干预结束后,各组大鼠联合灌胃给予非那西丁10 mg/kg、安非他酮10 mg/kg和奥美拉唑10 mg/kg并测定探针药给予前及给药后的0.083 h、0.25 h、0.5 h、1 h、1.5 h、2 h、2.5 h、3 h、4 h、6 h、8 h、12 h和24 h的血药浓度。结果与正常对照组相比,10 d组大鼠奥美拉唑AUC下降;清除率CL/F显著性升高;平均驻留时间MRT下降;10 d组大鼠安非他酮表观分布体积V/F显著性降低,上述差异均有统计学意义(P<0.05);探针药的其余药代参数差异均无统计学意义(P>0.05)。结论牙托水可诱导肝CYP2C19酶活性,而对CYP1A2与CYP2B6无影响。 Objective To elucidate the effects of methyl methacrylate on CYP1A2, CYP2B6 and CYP2C19 in rats. Methods 24 male SD rats were randomly divided into 3 groups: Control group, MMA-3d group and MMA-10d group. All rats of three groups were administered intragastrically with 10 mg/kg phenacetin, 10 mg/kg bupropion and 10 mg/kg omeprazole after administration of MMA for 3 or 10 consecutive days. Then, the blood samples were collected for analysis at 0, 0.083, 0.25,0.5, 1, 1.5, 2, 2.5, 3,4, 6, 8, 12 and 24 h after administration. Results Compared with the normal control group, the omeprazole AUC decreased in the 10 d group; the clearance rate of CL/F increased significantly; the mean residence time of MRT decreased; the apparent distribution volume of bupropion (V/F) in the 10 d group significantly decreased, and the above differences were statistically significant(P〈0.05 ). There were no statistical significance on the differences in the remaining pharmacokinetic parameters of the probe( P〉0.05 ). Conclusion The results maybe indicated that MMA had the potential to induce rat hepatic CYP2C19 activity, but not CYP1A2 and CYP2B6.
