摘要
目的对三峡库区沿江口岸的主要蚊类进行分子鉴定,分析各种蚊类的分子特征。方法用二氧化碳诱蚊灯在三峡库区沿江5个口岸采集成蚊,形态学鉴定之后,再提取DNA,扩增细胞色素C氧化酶亚基Ⅰ(COⅠ)基因并测序。将测序结果与GenBank中其他蚊类的DNA序列比对,并构建系统进化树。结果在宜昌、万州、涪陵、寸滩和泸州捕获三带喙库蚊、致倦库蚊、白纹伊蚊、骚扰阿蚊和中华按蚊共5个蚊种3 000余只,选取其中42只扩增COⅠ基因,获得5个蚊种的COⅠ基因序列,长度为658 bp,A+T含量为67.47%~69.71%。同源性比较表明,同一蚊种间核苷酸遗传距离小于0.0215,不同蚊种间遗传距离为0.0742~0.1594。COⅠ基因核苷酸系统进化关系显示,三带喙库蚊、致倦库蚊、中华按蚊、骚扰阿蚊和白纹伊蚊均聚集成簇,与形态学鉴定结果相一致。结论应用分子鉴定技术对三峡库区口岸蚊类进行鉴定,可为国境口岸蚊类鉴定提供溯源依据。
Objective To identify and analyze the molecular characteristics of the main mosquitoes in the ports along the Three Gorges Reservoir. Methods The mosquitoes were collected in the area of the ports along the Three Gorges Reservoir by using lamp traps,and morphological identified. Genome DNA was extracted and the cytochrome C oxidase subunit Ⅰ(COⅠ) gene was amplified and sequenced by universal primers. The phylogenetic tree was built based on COⅠgenes. Results A total of 3 000 mosquitoes represented 5 species were captured from Yichang,Wanzhou,Fuling,Cuntan and Luzhou. The CO Ⅰ gene of 42 mosquitoes were amplified successfully. The lengths of all of the COⅠ fragments were 658 bp,with A+T accounted for 67.5%-69.7%. Homological comparison showed that the nucleotide genetic distance of the same mosquito species was less than 0.0215,while that of different mosquito species was 0.0742-0.1594. Phylogenetic analysis showed that all of the mosquito species,such as Culex tritaeniorhynchus,Culex quinquefasciatus,Anopheles sinensis,Armiger essubalbatus,Aedes albopictus were gathered in their own clusters,which were consistent with morphological identification. Conclusion Molecular identification can be used to identify mosquito species effectively. This technique can help to identify mosquitos at frontier ports.
