摘要
目的:观察^(99)Tc–MDP对碱烧伤后大鼠角膜中基质金属蛋白酶2(MMP2)表达的影响。方法:清洁级成年健康的SD大鼠72只,制作碱烧伤模型后,分别按照0.005μg·5 mL^(-1)、0.01μg·5 mL^(-1)、0.05μg·5 mL^(-1)和等体积的0.9%氯化钠注射液予以局部滴眼。免疫组化染色后将染色结果采用病理图像分析仪分析MMP2含量,以平均光密度A(OD)值表示。结果:碱烧伤后不同浓度^(99)Tc–MDP给药组角膜MMP2表达有所下降,各组各时段与对照组相比,差异具有统计学意义(P<0.05)。结论:^(99)Tc–MDP滴眼可以下调碱烧伤后大鼠角膜内MMP2的表达。
Objective To observe the effects of ^99Tc-MDP on the expressions of matrix metalloproteinase-2 in the coneal of rats of alkali burn model. Methods 72 Sprague-Dawley rats were introduced with corneal alkali burn, with different concentrations, 0.005 ug/5 ml, 0.01 ug/5 ml, 0.05 ug/5 ml of ^99Tc-MDP and an equal volume of saline to drop eyes.Immunohistochemistry was used to examine the expression profiles of MMP2.The intensity of immunostaining of MMP2 was analyzed with a image analyzer and presented as averaged optical density（OD）. Results The corneal MMP2 expression in the group treated with different concentrations of ^99Tc-MDP was decreased after alkali burn, and there was a statistically significant difference between each group and the control group（P〈0.05）. Conclusion ^99Tc-MDP reduced the expressions of MMP2 in the cornea of rats of alkali model.
