摘要
目的探讨DLC-1基因在胰腺癌的发展过程中的作用。方法通过表达谱基因芯片分析和qPCR检测DLC-1基因在胰腺癌组织和癌旁组织中的表达差异,并检测不同胰腺癌细胞株中该基因的表达水平;同时过表达DLC-1基因的慢病毒感染胰腺癌细胞系SW1990,通过流式细胞术和Transwell实验观察DLC-1基因对胰腺癌细胞株的增殖及侵袭能力的影响。结果 DLC-1基因在胰腺癌组织中的表达显著低于癌旁组织;过表达DLC-1基因能够抑制胰腺癌细胞系SW1990的增殖和侵袭能力。结论 DLC-1基因的过表达对胰腺癌的发展有着明显的抑制作用。
Objective To investigate the role of DLC-1 gene in the development of pancreatic cancer. Methods The difference in expression of DLC-1 gene between pancreatic cancer tissues and adjacent tissues was detected,and the expression level of this gene in different pancreatic cancer cell lines was detected by qPCR and gene chip. Pancreatic cancer cells SW1990 were infected by lentiviruses with over-expressed DLC-1 gene. The effects of DLC-1 gene on the proliferation and invasive ability of the pancreatic cancer cell line were observed by FlowCytometry and Transwell test. Results The expression of DLC-1 in pancreatic cancer tissues was significantly lower than that in paracancerous tissues. Proliferation and invasion ability of pancreatic cancer cell line SW1990 were inhibited by overexpression of DLC-1 gene. Conclusion Overexpression of DLC-1 gene has a significant inhibitory effect on the development of pancreatic cancer.
