摘要
目的探讨白英生物碱对人肝癌Huh-7细胞增殖和凋亡的影响。方法 MTT法检测白英生物碱对Huh-7细胞增殖的影响,流式细胞术检测白英生物碱对人肝癌Huh-7细胞凋亡率的影响,蛋白印迹方法检测白英生物碱作用后细胞内凋亡相关蛋白的表达。结果白英生物碱能够抑制Huh-7细胞增殖,诱导Huh-7细胞凋亡,激活p38和Caspase-3蛋白。结论白英生物碱能诱导Huh-7细胞凋亡,其机制与调控p38和Caspase-3蛋白有关。
Objective To investigate the effects of Solanum lyratum Thunberg Alkaloid( STA) on the proliferation and apoptosis in Huh-7 cells invitro. Methods MTT assay was used to detect the effects of STA on the proliferation of Huh-7. The apoptosis was detected by flow cytometry. The expression of apoptosis protein was examined by Western blotting. Results STA could inhibit proliferation of Huh-7,induce apoptosis,and activate p38 and Caspase-3 protein. Conclusion STA could induce apoptosis of Huh-7,whose mechanism might be related with regulation of p38 and Caspase-3 protein.
