摘要
目的探讨miR-203表达与甲状腺乳头状癌发生与发展的关系。方法收集甲状腺乳头状癌标本40例,另选取良性甲状腺肿瘤标本30例,使用定量逆转录-聚合酶链反应技术(qRT-PCR)检测特定的miRNA表达,Transwell实验测定miR-203对细胞侵袭性的影响,通过启动子区乙酰化寻找与miR-203表达的关系。结果甲状腺乳头状癌组织及细胞系中miR-203的表达量均低于良性甲状腺组织或细胞系(P<0.05),且survivin的表达量有所上升(P<0.05);miR-203启动子区的去乙酰化会导致miR-203的表达异常。结论 miR-203在甲状腺乳头状癌中的表达低于甲状腺良性肿瘤组织,在甲状腺乳头状癌中的诊断及靶向治疗方面具有广泛应用。
Objective To investigate the relationship between the expression of miR-203 and the occurrence and de- velopment of thyroid papillary carcinoma. Methods Forty cases of thyroid papillary carcinoma and 30 cases of be-nign thyroid tumors were selected. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was used to detect specific miRNA expression. Transwell assay was used to determine the invasion of mi-203 cells. Acetylation in the promoter region was used to find the relationship with miR-203 expression. Results The expression of miR-203 in thyroid papillary carcinoma tissues and cell lines was lower than that in benign thyroid tissues or cell lines (all P 〈 0.05), and the expression of survivin was increased (all P 〈 0.05). Deacetylation of the promoter region leads to abnormal expression of miR-203. Conclusions The expression of miR-203 in papillary thyroid carcinoma is lower than that of benign thyroid tumor. It will be widely used in the diagnosis and tai^eted treatment of papillarythyroid carcinoma.
