摘要
目的探讨针刺内关、心俞穴对急性心肌梗死大鼠模型外周血血管内皮生长因子(VEGF)表达及心肌组织结构的影响。方法将72只雄性Wistar大鼠分成假手术组、模型组、内关组、心俞组、内关+心俞组、穴位旁组,每组12只。除假手术组外,其它5组建造急性心肌梗死模型。除假手术组和模型组,其他组均在建立大鼠心肌梗死模型后连续7d分别给予针刺相关穴位干预。7d后将各组大鼠处死并取材,用ELISA法检测各组大鼠血浆中VEGF的表达情况,电子显微镜观察除假手术组外5组左心室心肌损伤区心肌细胞的形态及心肌组织结构。结果血浆中VEGF表达,内关+心俞组>内关和心俞组>模型组及穴位旁组>假手术组,差异均有统计学意义(均P<0.05);内关组、心俞组、内关+心俞组的心肌细胞损伤程度要小于模型组及穴位旁组,其中内关+心俞组的心肌损伤程度最小。结论针刺内关穴、心俞穴可能通过上调VEGF的表达对心肌梗死后心肌组织的损伤具有保护作用。
Objective To investigate the effect of electroacupuncture at Neiguan and Xinshu on the expression of
vascular endothelial growth factor(VEGF)of the peripheral blood and the structure of myocardial tissue of rats with acute
myocardial infarction. Methods 72 male Wistar rats were divided into sham operation group, model group , Neiguan
group, Xinshu group, Neiguan + Xinshu group and beside acupoint group. After establishment of myocardial infarction
model in all groups aside from sham operation group, acupuncture was performed in Neiguan group, Xinshu group,
Neiguan + Xinshu group and beside acupoint group for 7 days. All rats were sacrificed and sampled 7 days later. The
expression of VEGF was measured by ELISA. The morphology of myocardial cells and the structure of myocardial tissue of
injured left ventricular myocardium were observed by electron microscope in all groups except sham operation group.
Results The expression of VEGF in the plasma were significantly higher in Neiguan + Xinshu group 〉 Neiguan and
Xinshu group〉model group and beside acupoint group〉 sham operation group(all P〈0.05). Myocardial cells damage
was significantly less in Neiguan group, Xinshu group and Neiguan + Xinshu group than in the model group and beside
acupoint group, and was smallest in Neiguan + Xinshu group. Conclusion Electroacupuncture at Neiguan point and
Xinshu point may reduce myocardial tissue damage after myocardial infarction by up regulating the expression of VEGF.
