PD-1在子痫前期患者外周血Treg/Th17细胞中的表达 被引量:5

Expression of PD-1 in Treg/Th17 cells in peripheral blood of patients with preeclampsia

摘要 目的研究子痫前期患者外周血中调节性T细胞(Treg)和辅助性T细胞(Th17)上程序性死亡受体-1(PD-1)的表达水平,探讨PD-1在子痫前期发病中的作用及意义。方法采集2016年在该院住院行剖宫产的单胎子痫前期孕妇外周血标本60例,其中轻度子痫前期(MP组)30例(MP组),重度子痫前期(SP组)30例(SP组);另选取同期在该院行正常剖宫产的孕妇30例作为对照组。采用流式细胞技术检测Treg细胞和Th17细胞百分比以及Treg和Th17细胞上PD-1的表达水平。结果 MP组和SP组外周血中Treg细胞较对照组明显降低(P<0.05),且SP组低于MP组(P<0.05),而Th17细胞结果与之相反,MP组和SP组较对照组明显升高(P<0.05),且SP组高于MP组(P<0.05)。与对照组相比,MP组和SP组患者外周血中Treg细胞表面表达PD-1百分比明显上调(P<0.05),且SP组高于MP组(P<0.05)。但是MP组和SP组患者外周血中Th17细胞表面表达PD-1百分比明显下降(P<0.05),且SP组低于MP组(P<0.05)。结论子痫前期患者外周血中Treg/Th17失衡,PD-1通过调控Treg/Th17参与子痫前期的发生。 Objective To research the expression levels of programmed cell death protein 1( PD-1) of regulatory T lymphocytes( Treg) and helper T lymphocytes( Th17) in peripheral blood of patients with preeclampsia,explore the role and significance of PD-1 in onset of preeclampsia. Methods In 2016,60 pregnant women with preeclampsia( single pregnancy) receiving cesarean section in the First People's Hospital of Aksu Prefecture were collected and divided into mild preeclampsia group( MP group) and severe preeclampsia group( SP group),30 women in each group. Thirty pregnant women receiving cesarean section in the hospital during the same period were selected as control group. Flow cytometry was used to detect the proportions of Treg cells and Th17 cells,the expression levels of PD-1 in Treg and Th17 cells. Results The levels of Treg cells in peripheral blood in MP group and SP group were statistically significantly lower than that in control group( P〈0. 05). The level of Treg cells in peripheral blood in SP group was statistically significantly lower than that in MP group( P〈0. 05). The levels of Th17 cells in peripheral blood in MP group and SP group were statistically significantly higher than that in control group( P〈0. 05). The level of Th17 cells in peripheral blood in SP group was statistically significantly higher than that in MP group( P〈0. 05). The proportions of PD-1 expressed on the surface of Treg cells in MP group and SP group were statistically significantly higher than that in control group( P〈0. 05). The proportion of PD-1 expressed on the surface of Treg cells in SP group was statistically significantly higher than that in MP group( P〈0. 05). The proportions of PD-1 expressed on the surface of Th17 cells in MP group and SP group were statistically significantly lower than that in control group( P〈0. 05).
