摘要
目的建立高效液相色谱法测定比格犬血浆中硝苯地平的含量。方法高效液相色谱法。色谱柱:C18色谱柱(4.6 mm×250 mm,5μm);进样量20μL;流动相为乙腈∶水=55∶45;流速1 m L·min-1;柱温30℃;检测波长237 nm。结果线性方程AS/AI=0.000 946X-0.002 187(r=0.999 2),线性范围(5~500 ng·m L-1),高、中、低3种浓度的硝苯地平测得的日内及日间精密度分别小于5.20%和3.59%,绝对提取回收率均大于71%,血浆中最低检测浓度为5 ng·m L-1。结论该方法操作简单方便,结果准确、稳定,可用于比格犬血浆中硝苯地平含量的测定和药动学研究。
Objective To establish a method for the determination of nifedipine in Beagle plasma.Methods Themethod involved liquid-liquid extraction and 20 μL of the resulting sample was analyzed by HPLC.Chromatographic separation was achieved on a C18 HPLC column (4.6 mm×250 mm,5 μm).The mobile phase was acetonitrile ∶ water = 55∶ 45 (V/V).The detection wavelength was 237 nm.The speed of flow was 1.0 mL·min^-1.Results The linear equation was AS / AI=0.000 946X-0.002 187( r = 0.999 2).The linearity of the method was established in the concentration ranges of 5 - 500μg·L^-1.The limit of detection was 5 μg·L^-1. Intra-day and Inter-day precisions were less than 5.20% and 3.59%,respectively.Conclusion This method can be applied to the determination of nifedipine plasma levels after administration ofcommercially available controlled-release nifedipine tablets to Beagle dogs.This method was simple,accurate and suitablefor pharmacokinetic studies of Beagle dogs.
