摘要
目的建立测定参附强心颗粒活性成分盐酸多巴胺的反相高效液相色谱法。方法样品用酸水溶解,0.45μm微孔滤膜过滤,采用JADE-PAK ODS-AQ-C18色谱柱(4.6 mm×250 mm,5μm)分析,以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速0.6 m L/min,柱温35℃,检测波长280 nm。结果盐酸多巴胺在0.0107~0.2138μg(r=1)与峰面积呈良好线性关系。平均加样回收率为100.8%,RSD为1.72%。结论本法准确、简单、重复性好,能更好地检测参附强心颗粒中盐酸多巴胺的含量。
Objective: To establish a reversed-phase high-performance liquid chromatography(RP-HPLC)method for the determination of dopamine hydrochloride in Shenfu Qiangxin Granules. Methods: The samples were dissolved with acid water,filtered with 0.45 um microporous membrane,and performed on a JADE-PAK ODS-AQ-C18 column with acetonitrile and 0.1% phosphoric acid solution as mobile phase in a gradient elution manner. The detection wavelength was set at 280 nm. Results: There was a good linear relationship between dopamine hydrochloride and peak area within the ranges of 0.0107~0.2138 μg(r = 1). Their average recoverie was100.8% and RSD was 1.72%. Conclusion: The method is accurate,simple and reproducible,and can better detect the content of dopamine hydrochloride in Shenfu Qiangxin Granules.
