摘要
目的:探讨谷胱甘肽硫转移酶A3(GSTA3)在鼻咽癌中的表达及其意义。方法:利用GCBI(Gene Cloud of Biotechnology Information)在线基因数据分析平台分析鼻咽癌原发肿瘤与正常对照组织的表达谱芯片数据(GSE13597、GSE64634、GSE12452),从中探寻GSTA3的表达水平。通过实时荧光定量PCR(qPCR)方法验证GSTA3在鼻咽癌细胞和组织中的转录水平差异,进一步利用免疫组织化学染色研究GSTA3在鼻咽癌肿瘤组织中蛋白表达的情况。结果:生物信息分析提示,GSTA3转录水平在3组鼻咽癌表达谱芯片中均表达下调(P<0.05);与正常组织比较,鼻咽癌组织中GSTA3mRNA的转录水平明显下降,差异具有统计学意义(P<0.05),与正常鼻咽上皮细胞NP69比较,5个鼻咽癌细胞系中GSTA3mRNA的转录水平均明显下降(均P<0.001);GSTA3蛋白在鼻咽癌患者中的表达水平低于正常组织(P<0.001)。结论:GSTA3在鼻咽癌中表达下调,可能参与鼻咽癌的发生和发展。
Objective:To investigate the expressions of Glutathione S-transferase A3(GSTA3)in nasopharyngeal carcinoma(NPC).Methods:Gene-Cloud of Biotechnology Information(GCBI)platform was used to analyze three cDNA microarray datasets(GSE13597,GSE64634,GS12452)of NPC,from which the expression of GSTA3 in NPC was found.The transcription of GSTA3 was detected in NPC cell line,NPC tissues,normal nasopharyngeal epithelial cell line and normal nasopharyngeal epitheliums(NNEs)by qPCR.The protein expression of GSTA3 was analyzed in NPC tissue chip(HNasC070 PG01)and NNEs by immunohistochemical staining.Results:Bioinformatic analysis data showed that the expression of GSTA3 was downregulated in NPC.We further confirmed that the expression of GSTA3 was down-regulated in NPC.Conclusion:The expression of GSTA3 was down-regulated in NPC,which might be the potential biomarker for diagnosis of NPC.
