摘要
目的:探讨温阳法中药复方对乳腺癌MCF-7细胞的生物学活性、细胞周期及抗侵袭转移的影响。方法:采用MTT法测定温阳法中药复方在不同作用时间下对乳腺癌MCF-7细胞的生长抑制作用;应用流式细胞技术检测其对细胞周期的影响;利用Western Blot法测定温阳法中药复方对乳腺癌MCF-7细胞肿瘤迁移和侵袭相关因子MMP-2及TIMP-2蛋白表达的影响。结果:倒置显微镜下观察发现,温阳法中药复方对人乳腺癌细胞MCF-7作用48 h后细胞发生不同程度的形态学改变;3600μg/mL剂量温阳法中药复方具有明显抑制乳腺癌MCF-7细胞的生长;1800、2700μg/mL剂量温阳法中药复方作用于乳腺癌MCF-7细胞48 h后,S期细胞数量显著增加;同时,1800、2700μg/mL剂量温阳法中药复方组MMP-2、TIMP-2的表达与空白对照组相比均有显著差异。结论:温阳法中药复方能够明显抑制乳腺癌MCF-7细胞生长,并且抑制作用具有时间-剂量依赖性,可将乳腺癌MCF-7细胞周期阻滞于S期,同时,通过调控MMP-2/TIMP-2平衡达到抗乳腺癌细胞侵袭转移的作用。
Objective: To research the block cell cycle and the influence of invasion and metastasis on breast cancer Cell MCF-7 by Yang-warming Formula. Method: We detected the growth of human breast cancer cell MCF-7 under the different doses of Yang-warming formula by the method of MTT. We measured the cell cycle of human breast cancer cell MCF-7 by the method of flow cytometry. Western blot was used for investigating the expressions of tumor metastasis related factors such as MMP-2 and TIMP-2 at the protein level. Result: Inverted microscope observation found different levels of morphological changes in the cell after 48 h of Yang-warming Formula on breast cancer cells MCF-7. The3600 μg/mL dose Yang-warming Formula has significant effect on breast cancer MCF-7 cell growth. S phase cells increased significantly after 48 h of using 1800 μg/mL and 2700 μg/mL Yang-warming Formula. At the same time,1800μg/mL and 2700 μg/mL dose Yang-warming Formula groups' expressions of MMP-2 and TIMP-2 were significantly different compared with those of the blank control group. Conclusion: Yang-warming Formula can obviously inhibit the growth of breast cancer MCF-7 cell,and with a time-dose-dependent inhibitory effect. It also can take the breast cancer MCF-7 cell cycle arrest in S phase,meanwhile,resist the invasion and metastasis of breast cancer cells through the regulation of MMP-2/TIMP-2 balance.
