摘要
目的探讨大鼠肝缺血再灌注损伤机制。方法建立SD大鼠肝缺血再灌注损伤模型,随机分为实验组(n=75)和对照组(n=75)。采用酶联免疫吸附试验(ELISA)法和Western blot法检测大鼠血清和肝脏组织白细胞介素-1(IL-1)表达水平和细胞间黏附分子-1(ICAM-1)、核因子-κB(NF-κB)的含量。结果(1)实验组和对照组肝缺血冉灌注后1、3、6h的IL-1的水平分剧为(90.45±14.76)、(101.19±10.84)、(116.77±13.87)pg/ml;(71.06±10.71)、(78.77±20.41)、(91.19±26.42)pg/ml。两组差异有统计学意义(P=0.000);(2)Western blot图谱分析,随着时间的延长,肝匀浆组织ICAM-1和NF-κB蛋白的含量逐渐增多,且实验组明显高于对照组。结论肝脏组织中IL-1、ICAM-1的表达增加与大鼠肝缺血再灌注损伤有关;其机制与通过激活NF-κB上调,对ICAM-1和IL-1表达调控加强有关。
Objective To study the mechanism of hepatic ischemiareperfusion injury in rats. Methods SD rat liver ischemia - reperfnsion injury model was established. The animals were randomly divided into experimental group and control group. The contents of serum interleukin - 1 ( IL - 1 ) and intereellular adhesion molecule - 1 ( ICAM - 1 ) and nuclear factor - κB ( NF - κB) in liver homogenate tissue were measured by enzyme linked immunosorbent assay (ELISA) and Western blotting. Results ( ] ) The levels of IL - 1 in the experimental group and the control group at 1 h, 3 h, and 6 h after operation were (90.45 ± 14.76), (101.19 ± 10.84), (116.77 ± 13.87) pg/ml, and (71.06 ± 10.71), (78.77 ± 20. 41 ) and (91.19 ± 26. 42 ) pg/ml respectively with the difference being significant between the two groups (P = 0. 000). (2) Western blotting analysis showed that the eontents of ICAM - 1 and NF - κB protein in liver homogenate tissue were gradually increased with time, and those in the experimental group were significantly higher than in the control group. Conclusion The increased expression of IL - 1 and ICAM - 1 in liver tissue is related to the hepatic ischemia - reperiusion injury in rats, which may be related to the up - regulation of ICAM - 1 and IL - 1 expression through activation of NF -κB.
