矾根‘欧布希迪昂’离体繁育技术研究初报 被引量:1

Preliminary Study on in vitro Breeding Technology of Heuchera micrantha ‘Obsidian’

摘要 矾根是近年来国内植物造景及家庭园艺中新兴的一种彩叶植物,在园林造景及城市彩化、美化中发挥着重要作用,但矾根种苗的生产繁育存在技术限制,无法满足市场的需求。本研究主要研究了矾根品种欧布希迪昂(Obsidian)的组培繁育技术。研究结果表明:75%乙醇消毒30 s+2%次氯酸钠消毒10 min为最适消毒方法。以MS为基本培养基,添加3 mg/L的6-BA和0.2 mg/L的NAA能够使矾根诱导率达90%;最适继代培养基是1.0 mg/L 6-BA溶液和0.1 mg/L NAA的混合培养基。2%次氯酸钠溶液是矾根工厂化育苗中外植体的最佳灭菌剂。 H. micrantha is a color-leafed plant emerging in plant landscaping and home gardening in China in recent years, and plays an important role in garden landscape and city beautifying, but the seedling production and breeding of H. micrantha are limited by technology, and cannot meet the demand of the market. This research mainly studied the tissue culture and breeding technology of H. micrantha varieties of European Bush Duhem(Obsidian). The results showed that 75% ethanol disinfection 30 s +2% 10 min sodium hypochlorite disinfection was the most suitable disinfection method. With MS as the basic medium, the inductivity of H. micrantha could reach 90% by adding 3 mg/L 6-BA and 0.2 mg/L NAA, and the optimal secondary medium was the mixed culture medium of 1.0 mg/L 6-BA solution and 0.1 mg/L NAA. 2% sodium hypochlorite solution is the best sterilization agent of explant in industrialized seedling production of H. micrantha.
