Effects of Alpha-GalCer on the Surface Maturation Markers Expression and Proinflammatory Cytokine Secretion of Lung Dendritic Cells in a Mouse Asthma Model

摘要 目的:观察α-半乳糖神经酰胺(α-GalCer)对哮喘小鼠肺树突状细胞(LDCs)表面分子和分泌细胞因子水平的影响。方法:将24只BALB/c小鼠随机分为正常对照组、哮喘组和哮喘α-GalCer组,每组8只。以卵清白蛋白(OVA)诱导哮喘动物模型,分别以α-GalCer或PBS干预。流式细胞术检测小鼠LDCs表面分子MHC-Ⅱ、CD40、CD86和CD80的表达水平,ELISA法检测小鼠LDCs培养上清液IL-12p70、IL-6、TNF-α和IL-10的水平;同时检测小鼠肺iNKT细胞数量和分泌细胞因子水平。结果:与哮喘组比较,哮喘α-GalCer组小鼠LDCs表面分子MHC-Ⅱ、CD40、CD86和CD80表达水平明显升高(P〈0.01),LDCs体外培养上清液IL-12p70、IL-6和TNF-α水平明显升高P〈0.01或P〈0.05);同时哮喘α-GalCer组小鼠肺iNKT细胞数量和分泌细胞因子水平明显增高(P〈0.01或P〈0.05)。结论:α-GalCer可以增强哮喘小鼠LDCs表面分子和促炎细胞因子表达水平,这可能与其活化肺iNKT细胞有关。 Objective:To investigate the effects ofα-GalCer on the surface maturation markers expression and cytokine secretion of lung dendritic cells(LDCs)in a mouse model of asthma.Methods:Twenty-four wild-type BALB/c mice were randomly divided into control group,asthma group(OVA-induced asthmatic mice treated with PBS),and asthmaα-GalCer group(OVA-induced asthmatic mice treated withα-GalCer).Flow cytometric analysis was used to detect the expression of MHC-Ⅱ,CD40,CD86,and CD80 of LDCs and the number of lung iNKT cells,IL-4+and IFN-γ+iNKT cells.Levels of proinflammatory cytokine in the culture supernatant of LDCs were measured by ELISA.Results:The expression of MHC-Ⅱ,CD40,CD86,and CD80 of LDCs fromα-GalCer group was markedly elevated,compared with those in asthma group(P〈0.01 or P〈0.05).The concentrations of IL-12 p70,IL-6,and TNF-αwere higher in the culture supernatant of LDCs inα-GalCer group than in asthma group(P〈0.01 or P〈0.05).Meanwhile,the number and activity of lung iNKT cells was significantly higher inα-GalCer group than those inasthma group(P〈0.01).Conclusion:α-GalCer can enhance the expression of surface maturation markers and secretion of proinflammatory cytokine of LDCs in a mouse asthma model,which maybe involve in activation of lung iNKT cells.
