枸杞子中甜菜碱HPLC-UV含量测定方法的优化 被引量:6

摘要 目的:优化枸杞子中甜菜碱的HPLC-UV含量测定方法。方法:采用甲醇提取,筛查碱性氧化铝与洗脱溶剂用量,优化前处理方法。采用XBridge Amide色谱柱(4.6 mm×250 mm,3.5μm),以乙腈-水(85∶15)洗脱;流速为1.0 mL·min^(-1);检测波长为195 nm;柱温30℃。结果:前处理制备方法优化为用甲醇加热回流提取,碱性氧化铝柱纯化处理。甜菜碱进样量在0.237 6~5.940 6μg范围内线性关系良好(r=0.9999),平均加样回收率(n=9)为97.42%(RSD=1.39%)。结论:该方法简便、可靠,重现性好,目标峰干扰少,可以用于枸杞子中甜菜碱的定量分析。经与现行药典方法比较,本法操作简便,测定准确性提高,可以用于现行标准的改进。 Objective:To optimize the determination method of betaine in Lycii Fructus by HPLC-UV. Methods: Methanol was used to extract the betaine. Alkaline aluminum oxide quantity and the elution solvent volume were screened to optimize the preprocess method. The test was performed in a XBridge Amide column (4. 6 mm×250mm, 3.5μm) under the isocratic elution of acetonitrile- water(85: 15). The flow rate was 1.0 mL·min^-1, the detection wavelength was set as 195 nm, and the column temperature was 30℃. Results:The optimized pre-process method was heating reflux extraction with methanol and purification with alkaline aluminum oxide column. The results showed the good linearity in the range of 0. 237 6 - 5. 940 6μg ( r = 0. 999 9 ) and the average recovery was 97. 42% with RSD as 1.39% (n = 9). Conclusion: The established method is simple, reliable and reproducible. It is suitable for the determination of betaine in Lycii Fructus as the less interference for the main peak, and can be referred to for the current method improvement as the simple operation and good accuracy compared to the Chinese Pharmacopoeia 2015 method.
