摘要 目的:探讨PTGER3在肠型胃癌中的表达及临床意义。方法:从GEO数据库的GSE29272数据集中下载58对肠型胃癌组织和癌旁组织的基因表达谱数据,筛选出差异表达基因,并利用DAVID数据库进行基因的功能富集分析。采用WGCNA软件对差异表达基因进行权重基因共表达网络分析,以挖掘出肠型胃癌发生发展过程中的关键调控基因。分析关键调控基因表达水平与肠型胃癌预后的相关性,并在Kaplan-Meier Plotter数据库中验证其预后意义。结果:共获得393个差异表达基因,DAVID功能富集分析显示它们主要涉及ECM受体相互作用、p53信号通路、PI3K-Akt信号通路和癌症信号通路等。PTGER3是癌症信号通路上的一个重要成员,在肠型胃癌组织中的表达明显上调。差异基因的共表达网络分析发现PTGER3是模块枢纽基因,与胃癌的发生发展关系密切。GSE29272数据集和Kaplan-Meier Plotter数据库的生存分析均显示PTGER3高表达与预后不良显著相关(P=0.034;P<0.001)。结论:PTGER3表达上调可能参与了肠型胃癌的发生发展,而且其高表达提示患者预后不良。 Objective:To explore the expression and clinical significance of PTGER3 in intestinal gastric cancer.Methods:The gene expression profile dataset GSE29272,which included 58 pairs of intestinal gastric cancer and adjacent tissues,was downloaded from GEO database.Differentially expressed genes were sorted,and a functional enrichment analysis was performed with DAVID.We further performed a weighted gene co-expression network analysis( WGCNA) to identify hub gene for the network.Survival analysis was conducted to assess the suitability of the hub gene identified as prognostic biomarker,and its prognostic value was further confirmed in the Kaplan-Meier Plotter database.Results:A total of 393 differentially expressed genes were identified.Functional enrichment analysis showed that these genes were mainly involved in ECM-receptor interaction,p53 signaling pathway,PI3 K-Akt signaling pathway and pathways in cancer.PTEGR3 was an important member of pathways in cancer,and was found significantly upregulated in intestinal gastric cancer.WGCNA revealed that PTGER3 was a module hub gene,which was associated with the development of gastric cancer.Kaplan-Meier analyses of two databases both showed that higher PTGER3 expression was significantly associated with worse patient prognosis( P = 0.034 and P〈0.001,respectively).Conclusion:PTEGR3 may plays an important role in the occurrence and development of intestinal gastric cancer.High PTEGR3 expression is associated with poor prognosis.
