石榴花多酚对离体大鼠胸主动脉环舒张作用的影响 被引量:5

摘要 目的研究石榴花多酚(Pomegranate Flower Polyphenol,PFP)对离体大鼠胸主动脉环舒缩功能的影响及作用机制。方法采用离体血管环灌流法观察(100~900)mg/L PFP对大鼠胸主动脉环张力的影响,给予钾通道阻断剂和一氧化氮合酶抑制剂N-硝基-L-精氨酸甲酯(L-NAME),测定大鼠血管组织中所含一氧化氮的含量。结果当PFP浓度为(700~900)mg/L时,与内皮去除组比较,内皮完整组PFP对大鼠胸主动脉环舒张作用更强,差异有统计学意义(P<0.01)。钾通道阻断剂格列本脲(Glib,10μmol/L)、四乙基氯化铵(TEA,3mmol/L)、氯化钡(BaCl_2,100μmol/L)预处理均能抑制PFP对内皮去除大鼠胸主动脉环的舒张效应。PFP对无钙高钾液中Ca^(2+)收缩曲线和无钙液中PE引起的内钙释放均有抑制作用。一氧化氮合酶抑制剂L-NAME预处理对PFP诱导的内皮完整大鼠胸主动脉环舒张作用具有抑制作用。结论石榴花多酚具有内皮依赖性和内皮非依赖性2种舒张血管作用。 Objective To investigate diastolic effect of Pomegranate Flower Polyphenol(PFP)on isolated rat thoracic aortic rings and its mechanism.Methods The tension of isolated thoracic aortic rings of rats in presence or absence of PFP(100~900mg/L)were observed by vascular ring perfusion method in vitro,then the mechanism were explored by application of Ca^(2+)K~+channel blockers and nitric oxide synthase inhibitor L-NAME.And the content of nitric oxide in rat vascular tissue was determined.Results When the concentration of PFP was(700~900)mg/L,PFP in the intact endothelium group had a stronger effect on rat thoracic aortic rings dilation than that in the group with endothelial deprivation(P〈0.01).Diastolic effect on denuded rat thoracic aortic rings induced by PFP could be inhibited by pretreatment of K~+channel blockers:10μmol/L Glib,3mmol/L TEA and 100μmol/L BaCl_2.PFP could move the calcium-constriction curve to the right in calcium-free high-potassium solution,and inhibit intracellular calcium releasing induced by PE in calcium-free solution.Pretreatment with nitric oxide synthase inhibitor L-NAME inhibited relaxation of thoracic aortic rings with intact endothelium induced by PFP.Conclusion PFP has endothelium-dependent and endothelium-independent vasodilation effects.
