摘要
研究香樟种子千粒重的变异水平,不仅有助于香樟的遗传改良,而且有助于香樟产业的发展。现有基于人工计数的香樟种子千粒重测量方法费时费力,而且容易出现误差,尤其不适合大样本的处理。该研究利用扫描仪获取标准图形,通过ImageJ软件自动计数,称重后得到千粒重,并与人工计数法进行了比较;利用该方法对江西境内具有地理代表性和古樟分布广泛的10个种源种子的千粒重进行了测定,分析了其种源及家系间的变异水平。结果表明:图像测定法能快速、准确对香樟种子进行计数,大大提高了千粒重测定的速度;江西局部种源香樟种子千粒重之间的变异主要来自于种源间,群体平均多样性指数为1.10,遗传多样性水平较低,且千粒重性状受地理气候因子影响较小。
The variation research on camphor 1000-seed weight will both contribute to the improvement of geneticbreeding and promote the development of camphor industry.However,the measurement of camphor 1000- seedweight based on artificial counting are time-consuming,laborious and prone to error,especially not suitable for largesample processing.In our research,we use the scanner to obtain the standard graphics,counting automatically by Im.ageJ software,calculating 1000-seed wright after weighing,and compared it with the artificial counting method.Thenthe camphor 1000-seed weight from Jiangxi provenance and its variation were analyzed.The results showed that scan.ner with ImageJ software provides a rapid and accurate way to the camphor seeds counting.1000-seed weight varia.tions among camphor trees from Jiangxi mainly come from the provenance,the average diversity index is 1.10 whichsuggested small genetic variation level among populations,and less affected by climatic factors.
