食管鳞状细胞癌组织中miR-21、PDCD4表达变化及意义 被引量:1

Expression changes and significance of miR-21 and PDCD4 in esophageal squamous-cell carcinoma tissues

摘要 目的观察食管鳞状细胞癌(食管鳞癌)组织中微小RNA-21(miR-21)、程序性细胞死亡基因4(PDCD4)表达变化,并探讨其临床意义。方法采用实时荧光定量PCR技术检测93例份食管鳞癌组织及癌旁组织中miR-21、PDCD4 mRNA表达,并以表达的平均值为界限,分别将食管鳞癌患者分为miR-21高表达组、miR-21低表达组及PDCD4 mRNA高表达组、PDCD4 mRNA低表达组。Spearman相关分析miR-21与PDCD4 mRNA表达的关系;Kaplan-Meier生存模型和Log-Rank检验分析miR-21、PDCD4表达对患者预后的影响。结果 miR-21在食管鳞癌组织中的相对表达量高于癌旁组织,PDCD4在食管鳞癌组织中的mRNA相对表达量低于癌旁组织,比较差异有统计学意义(P均<0.05)。经Spearman相关分析结果显示,食管鳞癌组织中miR-21与PDCD4的表达呈负相关(r=-0.883,P=0.000)。miR-21高表达组5年生存率低于miR-21低表达组(χ2=4.720,P=0.030),生存时间短于miR-21低表达组(P=0.029)。PDCD4 mRNA高表达组5年生存率高于PDCD4 mRNA低表达组(χ2=7.871,P=0.005),术后生存时间长于PDCD4 mRNA低表达组(P=0.006)。结论 miR-21在食管鳞癌组织中呈高表达、PDCD4呈低表达;二者可能参与食管鳞癌的发生发展,并影响患者预后。 Objective To observe the expression changes of miR-21 and programmed cell death 4( PDCD4) in the esophageal squamous-cell carcinoma tissues and to explore the clinical significance. Methods The real-time PCR was employed to detect the mRNA expression of miR-21 and PDCD4 in the esophageal squamous-cell carcinoma tissues and adjacent normal tissues. According to the average of the expression,the patients with the esophageal squamous cell carcinoma were divided into the high-miR-21 group and the low-miR-21 group,or the high-PDCD4 group and the low-PDCD4 group.The correlation between the miR-21 and PDCD4 in the esophageal squamous-cell carcinoma tissues was analyzed by the Spearman. The Kaplan Meier and the Log Rank were employed to detect the influence of miR-21 and PDCD4 expressions on the prognosis. Results The relative expression level of miR-21 in the esophageal squamous-cell carcinoma tissues was higher than that in the adjacent normal tissues,while the mRNA level of PDCD4 in the esophageal squamous-cell carcinoma tissues was lower than that in the adjacent normal tissues,and there were significant difference( both P 0. 05). The Spearman results showed that the miR-21 and PDCD4 was negatively correlated in the esophageal squamous-cell carcinoma tissues( r =-0. 883,P = 0. 000). The 5-year survival rate in the high-miR-21 group was lower than that in the lowermiR-21 group( χ^2= 4. 720,P = 0. 030),the same was the survival time( P = 0. 029). The 5-year survival rate in the high-PDCD4 group was higher than that in the low-PDCD4 group( χ^2= 7. 871,P = 0. 005),the same was the survival time( P = 0. 006). Conclusions In the esophageal squamous-cell carcinoma tissues,the miR-21 is highly expressed and the PDCD4 is low expressed,both of which may be involved in the occurrence and development of esophageal squamous-cell carcinoma,and have important influence on the prognosis.
