摘要
病毒入胞是病毒入侵的第一步。本研究前期已经率先发现人偏肺病毒存在胞吞的入胞方式。病毒的胞吞过程需要经历粘附,胞吞体和膜融合的过程。病毒入胞是病毒感染的第一个关键步骤,因此如果可以找到人偏肺病毒病毒胞吞入胞发生的关键时间段,不仅对于其入胞机制的研究,还有抗病毒策略的设计都大有帮助。本研究在研究前期基础上,通过全内反射荧光显微技术、激光共聚焦技术、R18/DiOC双波长成像技术等形态学的观测方式建立了一套观测病毒入胞时间段的方法。通过这套方法发现人偏肺病毒入胞的时间主要集中在感染后的前15 min。为了进一步验证入胞的关键时间段,本研究利用离心可以增强病毒感染的方法,验证了感染前15 min确实是人偏肺病毒入胞的关键时间段。本研究通过形态学观测的方法并验证发现人偏肺病毒入胞主要发生在感染后的前15 min,因此本研究将为今后人偏肺病毒入胞机制的阐明,抗病毒策略设计等提供关键信息,从而为疫苗和抗病毒药物研发提供理论基础。
Virus cell entry is the first step of invasion. Our group first found that human metapneumovirus enter into host cells by endocytosis. The endocytosis process of virus needs a process of attachment, vesicle transport and membrane fusion. Virus entry is the first critical step of virus infection, it will benefit to design anti-virus strategy if we found a critical time period of human metapneumovirus Cell Entry. In this study, total internal reflection fluorescence microscopy, laser confocal technique and R18/DiOC dual wavelength imaging technique were used to observe the process of hMPV entry, and we set a method to observe the time of hMPV cell entry.Through these methods, we found that the time of h MPV cell entry was mainly concentrated in the first 15 minutes after infection. In order to verify the critical time period of h MPV cell entry, we strength h MPV infection by centrifugation at different times, and the results showed that centrifugation at first 15 minutes significantly increase the virus infection. Therefore, we verified that hMPV cell entry occurred at the first 15 minutes after infection through the morphological observation methods. This study will provide a theoretical basis for the future development of vaccine and anti-hMPV drug.
