摘要
目的研究小柴胡汤水煎液对过氧化氢诱导的人皮肤成纤维细胞氧化应激损伤的保护作用。方法培养人皮肤成纤维细胞(HDF),建立HDF细胞H_2O_2损伤模型;使用不同浓度(20、50、100μg/ml)小柴胡汤水煎液干预H_2O_2损伤的HDF细胞,并设置H_2O_2模型组;MTT法检测细胞活力;化学发光法检测细胞内丙二醛(MDA)、超氧化物歧化酶(SOD)及活性氧(ROS)含量,蛋白质印迹法检测Bcl-2、Bax、NF-kB和p53蛋白表达。结果相对于H_2O_2模型组,20、50、100μg/L小柴胡汤水煎液保护组的细胞活力均显著提高(P<0.05;P<0.01;P<0.01);100μg/L柴胡汤水煎液保护组SOD活性显著升高(P<0.01),MDA含量和ROS荧光强度显著降低(P<0.01);100μg/L柴胡汤水煎液保护组NF-kB、p53、Bcl-2表达显著降低(P<0.05),而Bax表达却显著升高(P<0.05)。结论小柴胡汤水煎液能保护过氧化氢诱导的HDF细胞氧化应激损伤,其机制与降低胞内过氧化程度、阻断NF-kB通路有关。
Objective To explore the protectiveeffect of saikokeishitoin the oxidative damage of humanfibroblast cellsinduced by H2O2.Methods Humandermal fibroblast cells(HDF)were cultured and oxidative damage model induced by H2O2 was established,saikokeishito decoction with different concentrations(20,50,100μg/ml)was treated to HDF cell,and H2O2 model group was established,MTT method was used to detect cell viability,chemiluminescence was used to detect the content of malondialdehyde(MDA),superoxide dismutase(SOD)and reactive oxygen species(ROS);western blot was used to detect the expression of Bcl-2,Bax NF-kB and p53.Results Compared with H2O2 model group,cell viability of saikokeishito decoction groups with different concentrations(20,50,100μg/L)were significantly increased(P〈0.05,P〈0.01,P〈0.01),the activity of SOD in 100μg/L saikokeishito decoction group was significantly increased(P〈0.01),the content of MDA and the intensity of ROS were significantly decreased(P〈0.01),the expression of NF-kB,p53,Bcl-2 in 100μg/L saikokeishito decoction group were significantly decreased(P〈0.05),while the expression of Bax was significantly increased(P〈0.05).Conclusion Saikokeishito decoction can protect HDF cells from oxidative damage induced by H2O2,the mechanism is the reducing of intracellular oxidation andthe blocking of NF-kB pathway.
