摘要
目的建立高效液相色谱(HPLC)法同时测定通脉滴丸中葛根素、阿魏酸及丹酚酸B的含量。方法采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm);以甲醇-0.1%磷酸溶液为流动相,梯度洗脱;流速:1.1 m L·min-1;进样量:20μL;柱温:30℃;检测波长:250,321,286 nm。结果葛根素、阿魏酸、丹酚酸B的线性范围分别为41.6~416μg·m L-1(r=0.999 9)、3.84~38.4μg·m L-1(r=0.999 8)、36.8~368.0μg·m L-1(r=0.999 8);平均加样回收率分别为99.49%,99.73%,100.18%;RSD分别为1.11%,1.22%,0.94%。结论该法操作简单、结果准确,重复性高,可用于通脉滴丸中葛根素、阿魏酸和丹酚酸B的含量测定。
Objective To establish a HPLC method for simultaneous determination of puerarin,ferulic acid and salvianolic acid B in Tongmai dropping pills. Methods The Kromasil C18 chromatographic column（250 mm × 4. 6 mm,5 μm） was used,and the Column temperature was set at 30 ℃. The mobile phase was methanol-0. 1% phosphoric acid with a gradient elution. The flow rate was 1. 1 m L·min-1. The injection volume was 20 μL. The UV detection wavelength was set at250 nm,321 nm and 286 nm. Results The linear ranges of puerarin,ferulic acid and salvianolic acid B were 41. 6-416 μg·m L-1（r = 0.999 9）,3.84-38.4 μg·m L-1（r= 0.999 8）,36.8-368 μg.m L-1（r= 0.999 8）,respectively. The average recovery rates were 99.49%,99.73%and 100.18%,and RSDs were 1.11%,1.22% and 0.94%,respectively. Conclusion The method is simple,accurate,reproducible,and proper for simultaneous determination of puerarin,ferulic acid and salvianolic acid B in Tongmai dropping pills.
