摘要
目的:通过研究黄芪破壁饮片与传统饮片5种有效成分(黄芪甲苷、毛蕊异黄酮葡萄糖苷、芒柄花苷、毛蕊异黄酮、芒柄花素)在大鼠体内的药代动力学规律,进而探讨两者生物利用度的差异。方法:将大鼠随机分为两组灌胃给药:黄芪传统水煎液组和黄芪破壁饮片混悬液组(0.35 g·100 g);于不同时间点采集血样,血浆经蛋白质沉淀法处理后进行UPLC-MS/MS分析,测定血浆中5种成分浓度,DAS 3.2.6软件处理数据。结果:建立了测定大鼠血浆中5种有效成分的UPLC-MS/MS分析方法,其线性方程、稳定性、重复性、精密度及回收率均符合生物样品分析方法的要求。5种有效物质药时曲线均出现双峰现象,黄芪破壁饮片较黄芪传统饮片5种有效成分生物利用度提高1.4-2.5倍。结论:实验结果表明黄芪破壁饮片混悬液与黄芪传统饮片在大鼠血浆中有相似的代谢行为,黄芪破壁饮片混悬液较黄芪传统饮片生物利用度有所提高。
This paper was aimed to study the pharmacokinetic regularity of five effective components(Astragaloside A,Calycosin-7-glucoside, Ononin, Calycosin, Formononetin) in Ultrafine Granular Powder of Herbal Medicine(UGPHM)and Decoction Pieces(DP) of Astragali Radix(RA) in rats, so as to discuss the difference of bioavailability between UGPHM and DP. Rats, which were randomly divided into two groups, had intragastric administration of UGP and DP at the dose of 0.35 g ? 100 g. Blood samples were collected at different time points and purified by protein precipitation.UPLC-MS/MS was adopted to determine concentrations of five effective components, which were analyzed by DAS 3.2.6software. The results showed that the UPLC-MS/MS analysis method was adopted to determine five effective components in rat plasma, including linear equation, stability, repeatability, precision and recovery. The established sample processing and analysis methods were stable and reliable. The curve of five effective components of both the UGPHM and DP had bimodal effect. The bioavailability of five effective components of UGPHM of RA were 1.4-2.5 times comparing with DP of RA. It was concluded that according to experimental results, UDPHM and DP of RA had similar metabolic behavior, and UDPHM of RA had highly bioavailability comparing with DP of RA.
