摘要
目的:通过对AS家兔血管成形术后再狭窄炎性因子指标的分析,探讨通脉颗粒防治RS的机制。方法:雄性健康新西兰大白兔40只,随机分为空白组、模型组、辛伐他汀组、通脉颗粒组,每组10只。通过喂养高脂饲料建立家兔RS模型,成功建立冠状动脉再狭窄的模型,通过给药4周后,观察内膜增生及炎性因子之间的表达。通过组织病理学技术观察RS家兔髂动脉狭窄水平;通过免疫组化法观察RS模型家兔血管内MMP-2和TIMP-1的基因表达情况;采用RT-PCR方法检测损伤髂动脉MMP-2和TIMP-1的m RNA表达。结果:通脉颗粒组与模型组比较,能使MMP-2的表达明显下调,与此同时使TIMP-1表达上调(P〈0.05),与模型组对照,能明显下调MMP-2m RNA同时上调TIMP-1m RNA的m RNA表达(P〈0.05),并且与辛伐他汀组比较二者差异不显著(P〉0.05)。结论:通脉颗粒通过对炎性因子MMP-2和TIMP-1之间的表达,降低VSMC的迁移、增殖,起到了防治PTCA术后的再狭窄作用。
Objective :Through the analysis of the inflammatory factors of restenosis after angioplasty in AS rabbits,the mechanism of the prevention and treatment of RS by Tongmai Granule was discussed. Methods :Healthy male New Zealand white rabbit only 40 cases,were randomly divided into normal group,model group,simvastatin group,Tongmai Granule group,each group of 10 cases. The models of rabbits RS were established by feeding high-fat feed,and the models of coronary restenosis were successfully established. After 4 weeks of administration,the expression of intimal hyperplasia and inflammatory factors were observed. Through histopathological technique,we observed the stenosis degree of RS model rabbits. The gene expression of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-1(TIMP-1)in RS models were detected by immunohistochemistry. m RNA expression of MMP-2 and TIMP-1 was detected by RT-PCR. Results :Tongmai Granule group compared with model group,could significantly lower MMP-2 protein expression and increase TIMP-1 protein expression(P〈0.05),compared with model group,could obviously lower the expression of MMP-2 m RNA and also raised TIMP-1 m RNA expression(P〈0.05),Tongmai Granule group and simvastatin group had no significant difference(P〉0.05). Conclusion :Tongmai Granule can reduce the migration and proliferation of VSMC through the expression of MMP-2 and TIMP-1 of inflammatory cytokines,and play a role in the prevention and control of PTCA.
