血细胞分析仪XE-5000与流式细胞仪FACSCalibur在测定造血干细胞中的作用 被引量:2

摘要 目的探讨Sysmex XE-5000全自动血液分析仪和FACSCalibur流式细胞仪在测定造血干细胞中的相关性。方法分别应用XE-5000全自动血细胞分析仪上的HPC分析模式和流式细胞术对全血、外周血造血干细胞(PBSC)及白血病患者骨髓中的HPC及CD34+含量进行检测。结果测定的样本为自体动员的全血时,XE-5000血细胞分析仪测定的HPC绝对数与流式细胞仪测定的CD34+细胞绝对数呈正相关(r=0.52)。测定的样本为PBSC时,对于稀释5倍的PBSC样本其相关系数r=0.057。标本批内变异系数(CV)为3.9%。同一患者自体干细胞动员后的第4天WBC、CD34+和HPC同时明显上升,动员第5天同时达到高峰。结论在自体外周血干细胞移植时,流式细胞仪能准确测定造血干细胞的含量,Sysmex XE-5000虽然具有快捷、简捷的特点,但仍不能替代流式细胞术测定造血干细胞。
