摘要
目的探讨佳木斯地区汉族人群亚甲基四氢叶酸还原酶(methylenetetrahydrofolate reductase,MTHFR)C677T基因多态性与2型糖尿病(T2DM)患者血脂水平的关系。方法选取黑龙江省佳木斯地区汉族人群T2DM患者200例(T2DM组)及健康体检者150例(正常对照组),提取其血液样本DNA,PCR扩增目的基因后应用直接测序法对C677T多态位点分别进行基因分型,分析C677T多态性与T2DM患者血脂水平的相关性。结果 T2DM组CT、TT基因型及T等位基因频数与对照组比较有显著差异(P<0.01)。正常对照组T基因携带者与CC基因型相比,TG水平有显著性差异(P<0.01),且有更高的TC和LDL-C水平;T2DM组T基因携带者与CC基因型相比TC和LDL-C水平有明显差异(P<0.05),且有更高的TG水平。结论佳木斯地区汉族人群MTHFR C677T基因多态性可能与T2DM患者血脂水平相关。
Objective To investigate the relationship between MTHFR C677 T gene polymorphism and blood lipid levels in patients with type 2 diabetes mellitus（ T2DM） in Han population in Jiamusi region of Heilongjiang province. Methods 200 cases of T2DM patients（ T2DM Group） and 150 healthy cases（ Control group） in Han population in Jiamusi region of Heilongjiang province were selected.The DNA of the blood sample was extracted,the target gene was amplified by PCR,and the C677 T polymorphic loci were genotyped by direct sequencing.The association between C677 T polymorphism and blood lipid levels in T2DM was analyzed.Results The frequency of CT,TT genotypes and T alleles in the T2DM Group was significantly different from that of the control group（ P〈 0. 01）. Compared with the CC genotype,the T gene carriers in the Control group had statistically significant TG levels（ P〈 0.01） and higher levels of TC and LDL-C.The TC and LDL-C levels were statistically significant in T gene carriers in T2DM Group（ P〈 0.05）,and had higher levels of TG.Conclusions The gene polymorphism of MTHFR C677T may be associated with blood lipid levels of T2DM in Han polulation in Jiamusi region of Heilongjiang province.
