矾冰纳米乳对兔深Ⅱ度烧伤模型微血管密度及VEGF表达的影响 被引量:2

Effect of Alum-Borneol Nanoemulsion on Vascular Density of Rabbits with Deep Ⅱ Degree Burn

摘要 目的观察矾冰纳米乳对兔深Ⅱ度烫伤模型中微血管密度(MVD)及血管内皮生长因子(VEGF)表达的影响。方法将200只兔随机分为生理盐水组、烫疮油组、矾冰纳米乳低,中,高剂量组(8.15,16.3,32.6mg·m L^(-1)),每组40只。采用热力烫伤法造模,造模成功后2 h开始给药,每日2次,每次5 m L,分别于第3、7、14、21、28天,每组随机抽取8只大白兔处死,观察创面愈合情况并采集标本,免疫组化法检测MVD及VEGF的表达。结果矾冰纳米乳中剂量组MVD在第3,7,14和21天及VEGF在各个时间点与其余各组比较均优于各组,差异有统计学意义(P<0.05),实验各组MVD及VEGF与生理盐水组比较,差异均具有统计学意义(P<0.05),烫疮油组的MVD在第7和14天时间点与矾冰纳米乳低剂量组和矾冰纳米乳高剂量组比较,差异具有统计学意义(P<0.05);结论矾冰纳米乳能明显提高深Ⅱ度烧伤创面组织的MVD及VEGF的表达,促进烧伤创面的愈合,以中剂量组疗效最优。 Objective To observe the effect of alum-borneol nanoemulsion on the expression of microvessel density(MVD)in rabbits with deep second degree burn. Methods Two hundred rabbits were randomly divided into normal saline group,hot sore oil group,low dose group(8.15 mg·m L^-1),middle dose group(16.3 mg·m L^-1),high dose group(32.6 mg · m L^-1), 40 in each group. The rabbits were treated with hot scald method. HE staining was conducted to identify the success of modeling. After 2 hours,drugs were dispensed at the burn site twice per day with a volumn of 5 m L every time. After continuous administration,8 rats were sacrificed at the 3 rd,7 th,14 th,21 st and 28 th day respectively. The specimens were collected and the CD34 labeled MVD and VEGF were detected by immunohistochemistry. Results The MVD of the middle dose group was better than that of the other groups at the3 rd, 7 th, 14 th and 21 st day, and the VEGF was better than the other groups at all the time with statistically significant difference(P 〈 0.05). Compared with the saline group, the difference of MVD and VEGF of the experimental groups were statistically significant(P 〈 0.05). The MVD of the sore oil group was statistically significant(P 〈 0.05)at the 7 th and 14 thday compared with the low and high doses of alum-borneol microemulsion group. Compared with the sore oil group,there was no significant difference between VEGF of low dose group and high dose group(P 〉 0.05). The MVD and VEGF of the low dose and the high dose group showed no significant difference(P 〉 0.05). Conclusion Alum-borneol nanoemulsion can significantly improve the MVD and VEGF expression in deep second degree burn wounds,and promote the healing of burn wounds,among which the middle dose group(16.3 mg·m L^-1)showed the best effect.
