摘要 目的本研究通过构建IL-31RA基因敲除小鼠模型及哮喘模型,观察该基因敲除对哮喘小鼠气道炎症的影响。方法采用OVA建立小鼠哮喘模型,于末次激发后取肺组织HE染色及甲苯胺蓝染色观察肺部炎症细胞浸润和肥大细胞增生情况,支气管肺泡灌洗液(BALF)白细胞及嗜酸性粒细胞(EOS)计数,ELISA法检测血浆中总Ig E、IFN-γ、IL-4水平。结果成功构建IL-31RA基因敲除小鼠模型及哮喘模型,哮喘小鼠BALF中白细胞总数、嗜酸性粒细胞百分比和血浆中Ig E水平明显增多,基因敲除哮喘小鼠较野生型哮喘小鼠增多更明显。哮喘小鼠肺组织病理切片EOS、肥大细胞浸润,基因敲除小鼠较野生型小鼠EOS、肥大细胞浸润增多。哮喘小鼠血浆中Th2类细胞因子IL-4水平明显高于正常对照组,Th1类细胞因子IFN-γ水平明显低于对照组,且基因敲除哮喘小鼠IL-4水平较野生型哮喘小鼠增高。结论 IL-31受体A信号可抑制哮喘小鼠气道的炎症反应。 To investigate the effect of IL-31 receptor signaling on airway inflammation of mouse asthmamodel, an allergic asthmatic mouse model was induced by sensitizing and challenging with ovalbumin(OVA).Within 24 hours after last stimulating, the lung tissues were collected for HE staining and toluidine blue staining.The bronchoalveolar lavage fluid(BALF) were collected for WBC and EOS counting, while, the levels of Ig E, IFN-γand IL-4 in plasma were detected by ELISA. Data showed that the IL-31 RA gene knockout homozygous mousemodel and asthma mouse model had been established successfully. There were great increasing of total cells andeosinophils in the BALF of asthmatic groups; the levels of Ig E in KO-OVA group were significantly elevated, ascompared with control group, and were more significant higher compared with WT-OVA group. The inflammatorycell infiltration in BALF and lungs increased in KO-OVA group mice compared with WT-OVA group mice.Compared with the mice of control group, asthma mice displayed higher Th2 type cytokine level of IL-4, but lowerTh1 cytokine level of IFN-γ in plasma. Furthermore, the level of IL-4 in KO-OVA was significantly higher thanthat in WT-OVA group. Taken together, IL-31 receptor A signaling inhibits airway inflammation in asthma mice.
