摘要
为明确不同外源激素在油茶插穗生根过程的影响,获得高效油茶扦插外源激素配方,本研究以油茶插穗为材料,采用正交试验,研究不同外源激素配比对油茶插穗的成活率、生根率等形态建成指标的影响。结果表明:油茶插穗生根属于混合生根型,通过不同外源激素配比处理首次获得高达93.33%的混合生根率;不同外源激素配比处理的各类插穗生根形态建成指标均比清水对照高,不同外源激素配比处理间在生根率、混合生根率、愈伤组织面积和一级不定根数量中达到显著或极显著差异,而在成活率和愈伤组织产生率中差异不显著;经综合比较,认为处理A10(1 000 mg/L萘乙酸+3 000 mg/L吲哚丁酸+0.25 mg/L 2,4-D+1 mg/L复硝酚钠)效果最佳,成活率达100%、生根率最高达90.00%,混合生根率为86.67%,一级不定根数量为11.09条。极差分析表明,供试的四种外源激素对油茶扦插生根形态建成指标的影响差异不尽相同,萘乙酸和复硝酚钠在愈伤组织面积、生根率和混合生根率的影响比吲哚丁酸大;吲哚丁酸和复硝酚钠对一级不定根数量影响相当;萘乙酸对愈伤组织产生率的影响最大,其次是吲哚丁酸;吲哚丁酸对于成活率的影响最大。研究表明在植物扦插中不同外源激素的混合使用效果好,同时应重视复硝酚钠的使用。该研究为油茶扦插技术广泛应用于生产提供技术支持。
In order to clarify the effect of different exogenous hormones on rooting process of Camellia oleifera cuttings and obtain the formula of exogenous hormones, orthogonal experiment was used to study the effects of different exogenous hormones on the rooting of C. oleifera cuttings. The survival rate, rooting rate of C. oleifera cuttings under different ratio of exogenous hormones were studied. The results showed that the rooting of C. oleifera was a mixed rooting type, and the highest mixed rooting rate in this study was up to 93.33% for the first time. The rooting morphogenetic indexes under different exogenous hormone treatments were higher than that of water control.The rooting rate, mixed rooting rate, callus area and the number of primary adventitious roots was significant or extremely significant difference, but there was no significant difference in survival rate and callus formation rate between different exogenous hormone treatments. The best treatment was A10(1 000 mg/L naphthyleneacetic acid+3 000 mg/L indolebutyric acid +0.25 mg/L 2,4-Dichlorophenoxyacetic acid +1 mg/L sodium nitrophenolate), the survival rate, rooting rate, the mixed rooting rate and the number of primary adventitious roots of cuttings was the highest, 100%, 90%, 86.67% and 11.09 respectively. The range analysis showed that the effect of four exogenous hormones on rooting mor phogenesis of C. oleifera cuttings were significantly different. The effect of naphthaleneacetic acid and sodium nitrophenolate on callus area, rooting rate and mixed rooting rate were greater than that of Indolebutyric acid. The effect of indolebutyric acid and sodium nitrophenolate on the number of primary adventitious roots were similar. Naphthylacetic acid had the greatest effect on callus production, followed by indolebutyric acid. It was suggested that the effect of mixed use of different exogenous hormones was better than that of single hormone on rooting of plant cuttings, and sodium nitrophenolate should be pay attention to cuttings, the resear
