摘要
目的研究垂盆草苷对实验性幼年大鼠肝内胆汁淤积的干预作用。方法将60只幼龄SD大鼠随机分为空白组、α-荼基异硫氰酸盐(ANIT)组、熊去氧胆酸阳性对照组、垂盆草苷高、中、低剂量组,每组10只。除空白组外,其余各组大鼠均灌胃α-荼基异硫氰酸盐复制肝内胆汁淤积大鼠模型,各组大鼠于造模当日开始灌胃相应药物,每天1次,连续7 d。末次给药后测定大鼠血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)的活性和总胆红素(TBIL)的含量,检测肝匀浆中髓过氧化物酶(MPO)的水平。结果与空白组比较,ANIT组血清中ALT、AST、ALP、TBIL水平和肝匀浆中MPO活性升高(P<0.01);与ANIT组相比,各给药组大鼠血清中ALT、AST、ALP、TBIL水平和肝匀浆中MPO活性降低(P<0.05,P<0.01),垂盆草苷高剂量的作用与阳性药物作用相近。结论垂盆草苷对肝内胆汁淤积幼龄大鼠的肝功能具有保护作用。
Objective To study the protective effect of sarmentosin on liver function of young rats with acute intrahepatic cholestasis. Methods 60 young rats were randomly divided into blank control group,ANIT group,positive drug group,sarmentosin high-dose,middle-dose and low-dose groups,with 10 rats in each group. Those group were given α-naphthylisothiocyanate intragastrically to induce intrahepatic cholestasis model except blank control group. The rats of each group were given relevant medicine intragastrically on the day of modeling,once a day,for 7 day. The levels of alanine aminotransferase(ALT),alanine aminotransferase(AST),Alkaline phosphatase?(ALP),total bilirubin(TBIL) in serum and the activity of myeloperoxidase(MPO) in liver homogenate were detected in rats. Results Compared with blank group,the levels of ALT,AST,ALP,TBIL increased and the activity of MPO decreased in ANIT group(P〈0. 01). Compared with ANIT group,the levels of ALT,AST,ALP,TBIL decreased and the activity of MPO increased in each experimental group(P〈0. 05,P〈0. 01). The high-dose of sarmentosin has the same efficacy as Ademetionine 1,4-Butanedisulfonate? in the protection of liver function of rats with acute intrahepatic cholestasis. Conclusion Deoxyschizandrin have positive effect on liver function of intrahepatic cholestasis rats.
