柑橘渣酶解组合的筛选以及酶解条件优化 被引量:1

Screening of enzyme combinations of citrus pulp and optimization of its enzymolysis conditions

摘要 为了降低柑橘渣中的粗纤维含量、提高真蛋白和还原糖含量、改善饲喂价值,试验采用酶解方法进行酶解试验,设计了6组以筛选最佳酶解组合;随后研究酶组合比例、酶解温度、酶解时间以及料水比对柑橘渣酶解效果的影响。结果表明:果胶酶+纤维素酶+葡聚糖酶为最佳酶解组合;经过24 h酶解后,柑橘渣的物理结构发生明显改变,由致密变为疏松多孔状,粗纤维含量下降了7.03%,真蛋白含量提高了8.18%,还原糖含量提高了22.79%;柑橘渣酶解的最适比例为果胶酶、纤维素酶、葡聚糖酶之比3∶3∶2,最适温度为30~60℃,酶解时间为36 h,料水比为1∶2。说明在一定条件下采用果胶酶、纤维素酶和葡聚糖酶对柑橘渣进行酶解,可以有效地改善柑橘渣的营养价值。 The aim of the present study was to degrade the cellulose content and increase the pure protein and the reducing sugar level of citrus pulp,thus improve feeding value. Six enzymatic hydrolysis tests were performed by using enzymolysis method. Optimal enzymolysis combinations were screened. Then a series of experiments were performed to identify optimum enzymolysis conditions including the ratio of enzymes,enzymolysis temperature,enzymolysis time and the ratio of material to water. The results showed that the most effective enzyme combination was pectinase,cellulase and β-glucanase. After 24 h enzymolysis,the compact structure of citrus pulp turned into loose and porous. And the crude fibre content dropped by 7. 03%,pure protein content increased by 8. 18% and reducing sugar content added by 22. 79%. The results of optimal experiments showed that the best ratio of pectinase,cellulase,β-glucanase was 3 ∶3 ∶2,the optimal temperature was 30-60 ℃,the best hydrolysis time was 36 h,and the ratio of material to water was 1 ∶2. These results indicated that multi-enzyme combination of pectinase,cellulose andβ-glucanase could be used to enzymolysis citrus pulp under certain conditions which could improve nutrient value effectively.
