脾胃培源灌肠方对溃疡性结肠炎大鼠血清IL-17F含量的影响

摘要 目的:观察脾胃培源灌肠方对溃疡性结肠炎大鼠血清白介素-17F(interleukin-17F,IL-17F)含量的影响。方法:选用健康雄性SD大鼠120只,将120只SD大鼠随机分为:空白对照组、模型组、美沙拉嗪组、脾胃培源大剂量组、脾胃培源中剂量组、脾胃培源小剂量组,每组20只,采用TNBS/乙醇法复制UC大鼠模型灌肠诱导溃疡性结肠炎。造模成功后,空白对照组和模型组予3 m L生理盐水灌肠;美沙拉嗪组予美沙拉嗪悬浊液(0.36 g·L^(-1))3 m L·d-1灌肠;脾胃培源灌肠方大剂量组、脾胃培源灌肠方中剂量组、脾胃培源灌肠方小剂量组分别按含不同生药浓度的脾胃培源灌肠方3 m L·d-1(0.56 g·L^(-1)、0.28 g·L^(-1)、0.14 g·L^(-1))中药保留灌肠连续15 d,第16天处死大鼠,取结肠标本。观察大鼠结肠大体形态学改变情况,并采用ELISA法检测血清IL-17F含量。结果:模型组IL-17F含量较空白组显著升高(P<0.05);脾胃培源灌肠方组和美沙拉嗪组IL-17F含量较模型组显著降低(P<0.05),但两组间比较无统计学意义(P>0.05),但美沙拉嗪组和脾胃培源灌肠方大剂量组、脾胃培源灌肠方中剂量组、脾胃培源灌肠方小剂量组4组间两两比较,IL-17F含量无统计学意义(P>0.05)。结论:脾胃培源灌肠方能明显改善模型大鼠的结肠炎症损伤,抑制IL-17F的分泌。 Objective: To observe the influence of Spleen-Stomach Decoction for Edema to Strengthen Genuine Qi(SSDESGQ) on serum interleukin-17 F content of ulcerative colitis(UC) rats. Methods: One hundred and twenty 120 healthy male SD rats were selected and randomly divided into two groups: the blank control group,the model group,mesalazine group,SSDESGQ large dose group,SSDESGQ medium dose group,SSDESGQ small dose group,with 20 rats in each group. The rat model of UC was induced by replicating TNBS/ethanol method. After successful modeling,the rats in the blank control group and the model group were given 3 mL saline forenema. The ones in Mesalazine group were given mesalazine suspension(0. 36 g·L^-1) 3 mL·d^-1 for enema. The ones in the large,medium and small dosage groups of SSDESGQ were given 3 mL·d^-1 of SSDESGQ containing different concentration of raw medicine(0. 56 g·L^-1,0. 28 g·L^-1,0. 14 g·L^-1) for 15 consecutive days,and the rats were killed on the sixteenth day and colonic specimens were taken. The gross morphological changes of the colon in rats were observed,and the quantitative detection of serum IL-17F was performed by ELISA. Results: SSDESGQ can obviously improve the colitis injury in rats. The content of IL-17F in the model group was significantly higher than that in the blank group(P〈0. 05),and the contents of IL-17F in the SSDESGQ group and the mesalazine group were significantly lower than those in the model group(P〈0. 05),but there was no significant difference between the two groups(P〉0. 05). However,the difference in IL-17F content between any two of the mesalazine group and SSDESGQ large dose group,SSDESGQ medium dose group,and SSDESGQ small dose group had no significant difference. Conclusion: SSDESGQ can obviously improve the colitis injury and inhibit the secretion of IL-17F in the model rats. It has good curative effects on ulcerative colitis rats induced by TNBs/ethanol method.
