摘要
目的:优化松针总黄酮提取工艺,为合理利用东北松针资源提供理论依据。方法:以黑龙江省小兴安岭红松松针为原料,在单因素实验基础上,结合Box-Benhnken中心组合原理及响应面优化实验条件。结果:最优提取工艺参数:料液比为1∶65 g/mL,超声波功率比为70%,超声总时间为46 min,乙醇体积分数为55%,工作与间歇时间比为3/2(s/s)。该条件下松针总黄酮得率约为4.88%,为超声波细胞粉碎法提取松针黄酮的最佳工艺条件。结论:超声波细胞粉碎法更省时,可显著提高松针总黄酮得率,该方法为东北红松松针黄酮的提取提供了理论依据。
Objective:optimization of extraction process of total flavonoids in pine needles to provide theoretical basis for rational utilization of pine needles resources in Northeast China. Methods :using pine needles of Pinus koraiensis needle in Xiaoxing'anling of Heilongjiang province as raw materials,on the basis of single factor experiment,combined the principle of Box-Benhnken center combination and response surface optimization experimental conditions. Results :optimum extraction process parameters :the solid to liquid ratio is 1 ∶ 65 g/mL;the ultrasonic power ratio is 70%;the total time of ultrasound was 46 min;the ethanol volume fraction was 55%;the ratio of work to intermittent time is 3/2 s/s. Under these conditions,the yield of total flavonoids in pine needles was about 4.88%,thus,were the best conditions for extracting flavonoids from pine needles by ultrasonic cell pulverization. Conclusion :ultrasonic cell crushing method saves time and significantly improves the efficiency of flavonoids extraction from pine needles. The method provides a theoretical basis for the extraction of flavonoids from pine needles of northeast pine.
