两基原中药决明子中蒽醌类成分含量比较研究 被引量:5

摘要 目的:研究比较两基原中药决明子中蒽醌类成分含量,分析不同基源决明子产生非等效泻下作用的物质基础差异。方法:采用一测多评(QAMS)法,以大黄素为内参物,同时测定决明子中橙黄决明素、芦荟大黄素、大黄酸、黄决明素、大黄素、大黄酚、大黄素甲醚含量;采用紫外分光光度法(UV)测定决明子中游离蒽醌、结合蒽醌及总蒽醌含量;采用SIMCA-P11.5对含量测定结果进行PLS数理统计分析。结果:总蒽醌、游离蒽醌及结合蒽醌在决明中含量分别为1.41%~2.29%、0.21%~0.62%、1.51%~1.93%,在小决明中分别为0.77%~1.34%、0.13%~0.52%、0.52%~1.16%;PLS分析7种蒽醌类成分的VIP分别为1.15、1.12、0.69、0.90、1.21、1.15、0.52。结论:建立的QAMS方法重现性良好,测定的10批次决明样品中3批次大黄酚含量不合格,10批次小决明中6批次橙黄决明素不合格,7批次大黄酚不合格,决明中橙黄决明素、芦荟大黄素、大黄素、黄决明素含量均比小决明高;决明中总蒽醌、游离蒽醌及结合蒽醌含量均普遍高于小决明;决明中蒽醌类成分的差异主要由于大黄素、橙黄决明素、大黄酚、芦荟大黄素含量不同。 Objective:To comparatively study the content of anthraquinones in two-original TCM Cassiae Semen,and to analyze the substantial foundation diversity of purgation for different original Cassiae Semen.Methods:The QAMS was used to measure the content of aurantio obtusin,aloe emodin,chrysophanicacid,chrysoobtusin,emodin,chrysophanol,physcion simultaneously by setting emodine as a internal standard substance;UV was utilized to determine the content of combined,free and total anthraquinones in Cassiae Semen;SIMCA-P11.5 was used for PLS analysis with the data of content determination.Results:The content of combined,free and total anthraquinones in Semen of Cassia obtusifolia L.was 1.41%-2.29%,0.21%-0.62%,1.51%-1.93%;which in Semen of Cassia tora L.was 0.77%-1.34%,0.13%-0.52%,0.52%-1.16%.The result of PLS analysis showed that the VIP of seven anthraquinones was 1.15,1.12,0.69,0.90,1.21,1.15,0.52.Conclusion:The established QAMS method with good reproducibility was used to determine the content of seven anthraquinones.We found that the chrysophanol content in 3 of 10 batches of Cassia obtusifolia L.seeds and 7 of 10 batches of Cassia tora L.Seeds were unqualified,the Aurantio obtusin content in 6 of 10 atches of Cassia tora L.Seeds were unqualified.The content of aurantio obtusin,aloe emodin,emodin,chryso obtusin,even with total,free and combined anthraquinone in Cassia obtusifolia L.Seeds were generally higher than that of Cassia tora L.Seeds;The source of differentials for anthraquinones was the diversity content of Aurantio obtusifolin,emodin,chrysophanol and aloe emodin.
