摘要
通过对萃取剂选取、有机相体积比和反应温度的研究,对地衣芽孢杆菌发酵液中抗菌肽的萃取和反萃取条件进行了优化.结果显示,以正丁醇为萃取剂,V有机相/V水相为1∶2,操作温度为45℃时,单次萃取效率可达44.51%;设定V有机相/V水相为1∶1,操作温度为45℃时单次操作反萃取率为53.80%,二次操作反萃取率达61.28%.利用高效液相色谱对萃取液与反萃取液中的蛋白丰度进行检测,发现原发酵液中大部分的蛋白组分已被除去,从而达到了预处理的效果,有利于后续的分离工作.
The extraction and reversed extraction conditions of antimicrobial peptides in Bacillus licheniformis fermentation broth were optimizated by studying extraction agent selection,organic phase volume ratio and reaction temperature.The experimental results showed that with n-butanol as extractant,the single extraction efficiency rose up to 44.51% under operating temperature 45℃and volume ratio of organic phase and water phase 1∶1.When the operating temperature was 45℃,the single operation of the reversed extraction rate was 53.80%,the secondary operation of the reversed extraction rate reached61.28%.The protein abundance of the extraction and the reversed extraction solution were detected by high performance liquid chromatography.It was found that most of the protein components in the original fermentation broth had been removed,so as to achieve the effect of pretreatment,which was beneficial to the subsequent separation work.
