摘要
目的 探讨不同提取方法的金莲花汤提取物(金莲花汤不同提取物)对小鼠巨噬细胞Raw264.7增殖以及对小鼠H1N1流感病毒活性的影响方法 通过MTT法观察药物处理后对小鼠巨噬细胞Raw264.7生长增殖情况的影响;利用小鼠H1N1流感病毒感染MDCK细胞,进行病毒扩增并测定病毒效价;利用小鼠H1N1流感病毒感染小鼠巨噬细胞Raw264.7,采用MTT法结合CPE方法确定金莲花汤不同提取物的抗病毒效果。结果 MTT结果发现,金莲花汤不同提取物(20%、40%、60%、80%、95%、二次80%、二次95%及粗提取物)均对小鼠巨噬细胞Raw264.7增殖具有明显的抑制作用,IC_(50)分别为0.063、0.064、0.145、0.065、0.064、0.064、0.105、0.2mg/ml;MTT结果结合CPE分析显示,60%金莲花汤提取物IC_(25)及IC_(50)的药物剂量有轻微的抗病毒效果;80%金莲花汤提取物IC_0及二次95%金莲花汤提取物IC_(25)的药物剂量抗病毒效果较好,差异有统计学意义。结论不同提取方法的金莲花汤提取物对小鼠巨噬细胞Raw264.7增殖有一定的抑制作用;在体外抗病毒实验中,一定提取纯度的金莲花汤提取物对小鼠H1N1流感病毒有抑制作用。
Objective To investigate the effect of the soup of Trollius chinensis from different extraction methods on cell proliferation and the influenza virus(H1N1). Methods MTT method was used to observe cell proliferation in Raw264.7 treated with the extract of the soup of Trollius chinensis. We infected MDCK cells with the influenza virus(H1N1),and amplified the mice H1 N1 virus and detected the virus titer. MTT method combined with CPE was applied to detect the antiviral effect of the soup of Trollius chinensis from different extraction methods by infecting Raw264.7 with the influenza virus(H1 N1). Results MTT results showed that the soup of Trollius chinensis from different extraction methods had an obvious inhibitory effect on cell proliferation in Raw264.7 cells,and the IC_(50) was calculated and results were individually 0.063,0.064,0.145,0.065,0.064,0.064,0.105 and 0.2 mg/ml. MTT method combined with CPE results showed that 60% of the extract of Trollius chinensis soup had a slight anti-viral infection effect in IC_(25) and IC_(50) compared with viral infection group. Moreover,compared with viral infection group,the 80% of the extract of Trollius chinensis soup in IC_0 and the second extraction 95% of the extract of Trollius chinensis soup in IC_(25) had a much better antiviral role,and there were statistical significance. Conclusion The soup of Trollius chinensis from different extraction methods had an important effect on cell proliferation in Raw264.7. The extract of the soup of Trollius chinensis at a certain purity had anti-viral role on Raw264.7 cells infected by the influenza virus(H1N1) in vitro.
