摘要
药物成瘾是一种慢性复发性脑病,严重危害人类的身心健康,对社会公共卫生安全造成巨大威胁。因此,选择合适的动物模型,对药物成瘾的机制研究具有重要意义。目前,常用于成瘾研究的动物种属有大鼠、小鼠、猴等。其中非人灵长类动物在亲缘关系上与人最为接近,在生理、解剖及行为等方面与人类高度相似;其较长的生命周期也更适合长时程的药物成瘾研究,可以更好地模拟人类成瘾及复吸的病理进程,在成瘾研究中具有无可替代的重要地位。本文主要综述了目前常用的非人灵长类动物成瘾模型及其优势,重点介绍了非人灵长类动物在戒断试验、条件性位置偏爱试验、行为敏化试验及自身给药试验中的应用及特征,并对神经影像技术在此领域的应用作简要说明。
Drug addiction is a chronic,relapsing brain disorder,which not only induces a lot of serious body and psychological damages to addicts themselves,but also induces criminal activities with a threat to social and public safety.Therefore,it is important to select appropriate animal models for the study of the mechanism of drug addiction.At present,animal species commonly used in addiction research are rats,mice,monkeys and so on.Nonhuman primates(NHPs) are the closest relatives of humans,and the physiology,anatomy and behavior of NHP are more similar to humans than other laboratory species.Their long life span,well suited to long-term drug addiction research,can better simulate the pathological process of drug addiction and relapse in humans.In the study of addiction,NHPs have an irreplaceable important position.This article mainly reviewed the commonly used nonhuman primate models of drug addiction and their advantages,especially focused on the application and characteristics of nonhuman primates in the studies of withdrawal,conditional place preference,behavioral sensitization and self-administration,and briefly introduced the application of neuroimaging technology in this field.
