摘要
目的:通过复制脾气虚证和脾不统血证大鼠模型,对脾虚证由轻到重的发生发展过程的心肌组织能量代谢功能的变化进行探讨。方法:复制脾气虚证和脾不统血证大鼠模型,测定心肌组织ATP含量、柠檬酸合酶(CS)活性、线粒体呼吸链复合物(Complex Ⅰ、Complex Ⅱ、Complex Ⅲ、Complex Ⅳ)活性、线粒体DNA(mt DNA)拷贝数、COXⅣ蛋白和Cyt-C蛋白及其mRNA表达量变化。结果:脾气虚证大鼠和脾不统血证大鼠心肌ATP含量、CS活性、Complex Ⅰ、Complex Ⅱ、Complex Ⅲ和Complex Ⅳ活性、COXⅣ蛋白及其mRNA表达量均显著低于正常大鼠(P<0.05),脾不统血证大鼠mt DNA拷贝数、Cyt-C蛋白及其mRNA表达量显著高于正常大鼠(P<0.05);脾不统血证大鼠心肌CS活性、Cyt-C蛋白及其mRNA表达量显著高于脾气虚证大鼠(P< 0.05)。结论:大鼠脾气虚证早期,心肌组织能量代谢没有显著变化,随着脾虚病程加长及脾气虚证向脾不统血证的转变,大鼠心肌组织能量代谢功能开始发生障碍,ATP含量显著降低,三羧酸循环和电子传递链相关酶活性及其中重要蛋白和基因的表达都发生了改变,说明大鼠心肌组织在脾虚证发展加重的过程中受到了不利影响,符合中医脾气虚弱,则五脏无所滋养,积渐流于虚弱的理论观点。
Objective: To explore the developmental changes of energy metabolism in myocardium of the spleen deficiency from light to heavy dynamic process by replicating the rat models of spleen qi deficiency syndrome and spleen deficiency and failure to control circulating blood syndrome. Methods: The rat models of spleen qi deficiency syndrome and spleen deficiency and failure to control circulating blood syndrome were repli- cated. The levels of ATP, the activity of citrate synthase (CS), the activity of mitochondrial respiratory chain com- plex (Complex Ⅰ, Complex Ⅱ, Complex Ⅲ, Complex Ⅳ), the mtDNA copy number, the expression of COXIV and Cyt-C and their mRNA were measured in the myocardium of the rats. Results: The ATP content and activi- ties of CS, Complex Ⅰ, Complex Ⅱ, Complex Ⅲ, ComplexⅣ, expression of COXIV protein and its mRNA in rats with spleen qi deficiency syndrome and spleen deficiency and failure to control circulating blood syndrome were significantly lower than those in normal rats (P〈0.05). The mtDNA copy number and expression of Cyt-C protein and its mRNA in rats with spleen deficiency and failure to control circulating blood syndrome were significantly higher than those in normal rats (P〈0.05). The activities of CS and expression of Cyt-C protein and its mRNA in rats with spleen deficiency and failure to control circulating blood syndrome were significantly higher than those in spleen qi deficiency syndrome rats (P〈0.05). Conclusion: In the early stage of spleen qi deficiency syndrome in rats, there was no significant change in energy metabolism of the myocardium. Dysfunction of myocardial tis- sue energy metabolism in rats began to develop with the prolonged course of deficiency in spleen and the chang- ing from spleen qi deficiency syndrome to spleen deficiency and failure to control circulating blood syndrome.
