摘要
目的探讨E3泛素连接酶(CUL4A)蛋白在乳腺导管癌中的表达及其临床意义。方法应用免疫组织化学PV-6000法,检测15例癌周正常乳腺组织、16例中-高级导管原位癌组织、62例乳腺浸润性导管癌组织中CUL4A蛋白的表达。结果乳腺浸润性导管癌组织中CUL4A蛋白的表达阳性率明显高于中-高级导管原位癌组织、癌周正常乳腺组织(χ~2=24.143、6.806,P<0.05),乳腺浸润性导管癌组织CUL4A蛋白的阳性表达强度与腋窝淋巴结转移、TNM分期及组织学分级有关(Z=2.906~3.611,P<0.05),而与病人的年龄、肿瘤大小均无关(P>0.05)。结论 CUL4A促进乳腺导管癌的发生发展、浸润、转移,可能为抑制乳腺导管癌的浸润转移提供新的临床治疗靶标。
Objective To investigate the expression of E3 ubiquitin ligase CUL4 Aprotein in ductal breast cancer and its clinicopathological significance.Methods The PV-6000 immunohistochemical method was used to measure the expression of CUL4 Aprotein in 15 normal adjacent breast tissue samples,16 samples of intermediate-to-high-grade ductal carcinoma in situ(DCIS),and 62 samples of invasive ductal carcinoma(IDC).Results IDC tissue samples had a significantly higher positive rate of CUL4 Aprotein than intermediate-to-high-grade DCIS tissue samples and normal adjacent breast tissue samples(χ2=24.143 and6.806,P〈0.05).The expression intensity of CUL4 Aprotein in IDC tissue samples was associated with axillary lymph node metastasis,clinical stage,and histological grade(Z=2.906-3.611,P〈0.05),but it was not associated with patient's age and tumor size(P〈0.05).Conclusion CUL4 Apromotes the development,progression,invasion,and metastasis of ductal breast cancer,which may provide a new target for inhibiting the invasion and metastasis of ductal breast cancer.
