摘要
目的:观察绿茶提取物对口腔鳞癌模型大鼠抗肿瘤组织中Eph A2、Ephrin A1和E-cadherin表达的影响。方法:将45只健康Wistar大鼠分为正常对照组、模型对照组、绿茶提取物组,每组各15只,模型对照组以及绿茶提取物组给予5 g/L 4-NQO涂于口腔腭黏膜以建立口腔鳞癌模型大鼠,正常对照组不涂抹,正常饲养。绿茶提取物组于模型建立后开始给予绿茶提取物50 mg混合10 m L生理盐水,每日分2次灌胃,给药8周。其余2组给予生理盐水灌胃,剂量与使用频率和绿茶提取物组相同至实验结束。比较各组大鼠情况、组织中Eph A2、Ephrin A1和E-cadherin表达情况。结果:实验过程中严格遵守无菌操作,其中正常对照组无大鼠死亡,模型对照组大鼠4只死亡,死亡发生率为26.67%;绿茶提取物组1只死亡,死亡发生率为13.33%。治疗后,与正常对照组相比,模型对照组以及绿茶提取物组Eph A2、Ephrin A1平均光密度水平较高(P<0.05),E-cadherin平均光密度水平较低(P<0.05);与模型对照组相比,绿茶提取物组Eph A2、Ephrin A1平均光密度较低(P<0.05),E-cadherin平均光密度较高(P<0.05)。结论:绿茶提取物能够改善口腔鳞癌模型大鼠症状,阻止癌细胞生长,可能与下调癌症组织Eph A2、Ephrin A1表达水平,上调E-cadherin表达水平有关。
Objective: To observe the effects of green tea extract on the expressions of Eph A2, Ephrin A1 and E-cadherin in antitumor tissue of rat model with oral squamous cell carcinoma(OSCC). Methods: All 45 healthy Wistar rats were divided into normal control group, model control group and green tea extract group, 15 rats each group, the rats of model control group and green tea extract group were prepared to be OSCC rat model by smearing with 4-NQO, 5 g/L, over oral and palate mucosa, normal control group left unhandled and accepted normal feeding.The rats in green tea extract group received the lavage of green tea extract, 50 mg, mixing with physiological saline, 10 m L, twice each day, for eight weeks. The other two groups were drenched with physiological saline, the dose and the frequency were same to green tea extract group by the end of the experiment. The expressions of Eph A2, Ephrin A1 and E-cadherin in the tissue and the conditions of the rat model were compared. Results: Aseptic technique was strictly conducted in the experimental process, and there was no death in normal control group, four died in model control group, mortality rate reached 26.67%; one died in green tea extract group, and mortality rate reached 13.33%.After treating, compared with normal control group, average optical levels of Eph A2 and Ephrin A1 in model control group and green tea extract group were higher(P〈0.05), while average optical level of E-cadherin was lower(P〈0.05);compared with model control group, average optical levels of Eph A2 and Ephrin A1 in green tea extract group were lower(P 〈0.05), while average optical level of E-cadherin was higher(P〈 0.05). Conclusion: Green tea extract could improve the symptoms of OSCC rats, inhibit the growth of cancer cells, which might be related to down regulating the expressions of Eph A2 and Ephrin A1, up regulating the expressions of E-cadherin in cancer tissue.
