摘要
经驯化后,挑选鱼龄为36 d,初始体长为(8.02±0.32)cm、体重为(7.32±0.26)g的健壮军曹鱼稚鱼420尾随机分成7组,每组3个平行,每个平行20尾鱼,分别投喂7组饲料中的一种,这7组饲料分别是:对照组(D0),只含基础饲料,不添加VE和CA;实验饲料D1~D6,在每千克干基础饲料中分别添加VE 100、0、100、75、50、25 IU和CA 0、12、12、6、3、1.5 g,投喂12周,探究不同剂量维生素E(VE)和/或柠檬酸(CA)添加剂对军曹鱼(Rachycentron canadum)稚幼鱼主要组织/器官的顺乌头酸酶(ACO)和过氧化物酶体增殖物激活受体α(PPARα)基因的表达、抗氧化酶活力和多不饱和脂肪酸代谢的影响,为开发适合军曹鱼稚幼鱼配合饲料提供理论依据。结果显示:投喂添加不同剂量VE和/或CA日粮的军曹鱼的肝、肌肉、心脏、脑和肾ACO和PPARα基因表达量、SOD、CAT、GPx和GST的活力、PUFAs和n-3高度不饱和脂肪酸(n-3 HUFAs)含量均显著高于对照组(P<0.05)。投喂联合添加VE和CA日粮(D3~D6组)的鱼上述指标分别显著高于投喂单独添加VE(D1组)或CA(D2组)日粮鱼的相应指标(P<0.05)。D5组鱼的ACO和PPARα基因表达量、SOD、CAT、GPx、GST、PUFAs和n-3 PUFAs含量最高,分别显著高于其他组鱼的相应指标(P<0.05)。在本实验条件下,添加不同剂量的VE和/或CA添加剂能够显著提高军曹鱼稚幼鱼的ACO和PPARα基因表达量、抗氧化酶活力、PUFAs和n-3 HUFAs含量;联合添加VE和CA能够协同显著提高鱼的上述指标,分别显著高于单独添加VE(D1组)或CA(D2组)者;最佳添加量是VE 50 IU和CA 3 g/kg干饲料。
Four hundred and twenty of juveniles cobia with age of 36 d, initial body length of (8.02± 0.32) cm, body weight of (7.32±0.26) g were divided randomly into 7 groups, with 3 replicates per group and 20 fish in each replicate. The 7 groups of the juveniles were fed using one of 7 specific diets. The 7 diets were: one control diet, DO, which had no VE and CA added, and 6 experimental diets, D1-D6, to which were added VE 100, 0, 100, 75, 50, 25 IU and CA 0, 12, 12, 6, 3, 1.5 g per kg of dried feed, respectively. The juveniles were fed for 12 weeks to investigate the effect of dietary vitamin E and/or citric acid supplementation on the expression of ACO and PPARα genes, activi- ty of antioxidant enzymes, and PUFAs in juvenile cobia to develop a suitable artificial feed for cobia. The results showed that the expression of ACO and PPARα genes, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione-S transferase (GST), PUFAs, n-3 highly unsaturated fatty acids (n-3 HUFAs) in the analytical tissues/organs of cobia juveniles fed the different dietary VE and/or CA supplement were significantly higher (P〈0.05) than those of the fish fed DO, respectively. These parameters of the juveniles fed the diets with both VE and CA supplement (D3-D6) were significantly higher (P〈0.05) than those of the fish fed the diets with VE (D1) or CA (D2) supplement alone, respectively. The highest expression of ACO and PPARα genes, SOD, EAT, GPx, GST, PUFAs, and n-3 HUFAs were found in the liver, muscle, heart, brain, and kidney of the D5 fish and differed significantly (P〈0.05) from those of the fish fed other diets, respectively. Conclusions were drawn that different dietary VE and/or CA supplement could considerably promote the expression of ACO and PPARoL genes, the activity of antioxidant enzymes, and PUFAs in cobia juveniles; the combined dietary VE and CA supplement could cooperate and significantly promote above parameters; the optimum diet
